Effects Of Osteopontin On ADAMTS-4and ADAMTS-5of Chondrocyte In Patients With Osteoarthritis Of The Knee

Objective:The study is designed to change the contents of OPN in the chondrocytes, in order to observe the effect of OPN on the expression of ADAMTS-4and ADAMTS-5of chondrocytes in patients with osteoarthritis of the knee.Methods:The study was performed on16specimens of cartilage obtained from osteoarthritic patients who underwent total knee arthoplasty surgery. All the cartilage samples were cultured and purified to obtain chondrocytes in vitro. Taking the frist passaged cells into three groups:(1) Group A (the control group);(2) Group B (treated with100ng/ml OPN for48hours);(3) Group C (treated with1μg/ml OPN for48hours). Then design and validate the real-time quantitative polymerase chain reaction (Q-PCR) assays with SYBR Green dyes for simultaneous quantification of the expression of the ADAMTS-4mRNA and ADAMTS-5mRNA. All results statistics were analysed by SPSS19.0.Results:1. Compared to controls, OPN (100ng/ml) downregulated ADAMTS-4mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).2. Compared to controls, OPN (1μg/ml) downregulated ADAMTS-4mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).3. Compared to OPN (100ng/ml), OPN (1μg/ml) downregulated ADAMTS-4mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).4. There was a negative correlation between the expression of ADAMTS-4mRNA and the concentration of OPN in the three groups(r=-0.455,p<0.05).5.Compared to controls, after OPN (100ng/ml) interferenced, the expression of ADAMTS-5in OA chondrocytes, showed no difference of statistically significant(p>0.05).6. Compared to controls, after OPN (1μg/ml) interferenced, the expression of ADAMTS-5in OA chondrocytes, showed no difference of statistically significant(p>0.05).7. The difference of ADAMTS-5mRNA expression between OPN (100ng/ml) and OPN (1μg/ml) interferenced in OA chondrocytes, showed no difference of statistically significant(p>0.05).Conclusion:1. OPN can downregulate ADAMTS-4mRNA expression in OA chondrocytes, and there is a negative correlation between the expression of ADAMTS-4mRNA and the concentration of OPN increased in OA cartilages. 2. OPN can not affect ADAMTS-5mRNA expression in OA chondrocytes, and there is no relationship between the expression of ADAMTS-5mRNA and the concentration of OPN increased in OA cartilages.
