Objective: Osteoarthritis is one of the most common knee joint disease, although it is achronic knee joint disease, it will lead to disability with the progression of the Osteoarthritis,and the Osteoarthritis is a chronic, progressive, degenerative joint disease, articular cartilagedegeneration is the main characteristics, joint pain and joint dysfunction is the main clinicalmanifestations, it is a set of induced by different causes but with similar morphology, biologyand clinical manifestations of common disease. According to the result of epidemiologicalinvestigation, in the past ten years the world number of osteoarthritis patients have increasedby thirty percent, and the incidence of osteoarthritis in the world older than50years oldpeople is about fifty percent, in order to understand and study the pathogenesis and treatmentsof osteoarthritis, this study discusses and research the expression and correlation ofADAMTS-4and ADAMTS-5in osteoarthritis patients synovial.Methods: Collect the synovial specimens from the first affiliated hospital of ShiheziUniversity from March2012to March2013in our department (The first orthopaedic) treatedwith the surgery of the articular cavity mirror, knee joint replacement or lower extremityamputation surgery of60patients (experimental group choose30patients with clinicaldiagnosis of OA; control group choose pure knee meniscus injury, synovial plica syndrome oramputation patients,30cases),36male patients,24female patients, aged form18to75yearsold, the median age is60years old, and all cases were diagnosed by our hospital orthopedicand radiographic examination (using1995American rheumatism association about OAdiagnostic criteria), with the consent of the patients and hospital ethics committee approvedand then took samples. Using Western blot and immunohistochemical detection theexpression of ADAMTS-4and ADAMTS-5in the synovial of30patients with osteoarthritis(experimental group) and30cases of control group. The experimental data for statisticalanalysis using SPSS17.0software, the study counting data using chi-square inspection andexpressed by x±s, the expression of these two kinds of protein and the correlation relateddegree with OA using the Pearson product moment correlation analysis, r means correlationcoefficient, P <0.05is defined as have statistical significance in the difference.Results: Western blot results showed that the expression of ADAMTS-4/-5protein inosteoarthritis patients synovium of knee joint was significantly higher than those in non-osteoarthritis, the expression of ADAMTS-4were positively correlated with ADAMTS-5inOA synovium tissue. IHC results showed that the expression of ADAMTS-4and ADAMTS-5in OA patients synovium of knee joint was markedly higher than that in normal synovium(P value<0.01), and the ADAMTS-4and ADAMTS-5were mainly located in the cytoplasm ofsynovium cells.Conclusion: There is a related association between expression of ADAMTS-4/-5protein andthe progression of OA. They might be an important indicator in diagnosis and judgment ofosteoarthritis.
