Objective:To investigate the effects of activated platelets on the expression of LOX-1(Lectin-like oxidized LDL receptor-1) in human endothelium.Methods:This study was carried out at the Department of Cardiology in the Xiangya Hosptial, Central South University,China, from September2011to Jaunary2012.We collected55Acute Coronary Syndrome(ACS) patients. For the Healthy Control group,49healthy subjects were enrolled from Xiang ya medical center. For all the subjects, we gave them preformed questionnaires to collect data of weight and other basic information. We also collected blood samples from the vein, and measured the plasma LOX-1level using Enzyme linked immunosorbent assay.also The expression of CD62P and PAC-1in platelets were analyzed by flow cytometer. Furthermore, we extracted platelets from the blood samples and then incubated human umbilical vein endothelial cells HUVECs with the extracted platelets,.Measure the LOX-1level in the medium was measured using Enzyme linked immunosorbent assayResult:There was no statistically significant differeces in the age,systolic and diastolic blood pressure,FBS, Platelet number between ACS group and control group(P>0.05), but the triglyceride, high density lipoprotein, low density lipoprotein have statistically significant differeces.CD62P(25.72±6.64)%, PAC-1(29.05±7.76)%, LOX-1concentration (27.57±5.36pg/ml)in ACS group,CD62P(2.49±1.01)%, PAC-1(2.1±1.03)%, LOX-1concentration (24.51±3.16pg/ml) in Healthy Control group, the CD62P, PAC-1, LOX-1concentration in ACS group were higher than Healthy Control group, and the differences were statistically significant(P<0.05). And the expression of CD62P and PAC-1have a significantly correlated with plasma levels of LOX-1in ACS group(CD62P and LOX-1r=0.451, P<0.01;PAC-1and LOX-1, r=0.567,p<0.01), and the Healthy Control group do not have this relation(CD62P and LOX-1r=-0.274, P=0.065;PAC-1and LOX-1, r=-0.080, P=0.599).In the cell experiments, the LOX-1concentration in the medium of ACS group was21.45±6.15pg/ml, and the Healthy Control group is4.91±1.70pg/ml, the Control group is4.52±1.58pg/ml, the concentration in ACS group was higher than the Healthy Control group and Control group, and the difference between them was statistically significant.Compare the Healthy Control group and the Control group,the differences between them were no statistically significant.Conclusion:Plasma levels of LOX-1,and the CD62-1,PAC-1level in platelet were increased in the Acute Coronary Syndrome patients; The expression of CD62P and PAC-1in platelet have a correlation with plasma LOX-1levels in Acute Coronary Syndrome patients; Activated platelets can increase LOX-1expression in endothelial cells.
