The epidemiological data show that the high morbidity, mortality and mutilation rate made cerebrovascular diseases (CVD) one of the three main reason of death and greatly harmful to the health of human being. The intracerebral hemorrhage (ICH) is one of the most critical diseases for the urgent and various symptoms in acute stage. The treatment in the stage is important to the salvage and healing. Now apoptosis has been confirmed to be the main style of neuron death in delay injury of ischemia-reperfusion (IR). To suppress apoptosis can reduce the injury of nerve cells in edema band. The Chinese medicinal treatment is predonminant in this stage. In Chinese medical theory, the essential mechanism of stroke is the origin deficiency and excess in superficiality. The location is in brain. The brain protective effects and mechanism of Jiunaoning Inject ion (JNN) were studied by animal and cell culture experiments in this thesis that guided by Chinese medicinal theory and based on the neuron apoptosis technique. 1. The protection of JNN on ICH experimental rats ICI-I model was made by injecting minim collagenase VU. Male Wis tar rat, weighing 240?00g, were divided into the six groups of normal, control, INN (large, middle, small dosage) and Qingkailing Injection (QKIJ. JNN and QKL were administrated by intraperitoneal injection, and all of the experimental indicators were detected on the 3rd, 7th and 15th day. 1. 1 The effect of JNN on cerebral edema and nerves function of ICH experimental rats Results showed that cerebral edema in the groups of JNN and QKL decreased significantly (P<0. 01), especially in the group of INN of middle dosage. The symptoms of nerves function in the three dosage groups of INN improved remarkably (P<0. 05 or P<0. 01) 1.2 The effect of INN on brain tissue morphological changes of ICH experimental rats The brain tissue slices were stained by I-YE, Nissl and Massom stain for the immunohistochemistric studies. The control group was found that the neuronal morphologic changes in ICY-I included numerous necrosis of nerve cells and infiltration of inflammatory cells in hematoma region. Neuron lost partly, remained cells swollen, acidophilia change of cytoplasm, pyknosis of nucleus and reduction of Nissl body in remained cells and glial cell reaction in hippocampal CAl area. These changes in the three dosage groups of INN and QKL were smaller than that in the group of control on the 3rd, 7th and 15th day. These results indicated that INN could improve the absorption of hematoma and relieve cerebral edema. 2. The effective mechanisms of INN on nerve cell apoptosis of ICE-I experimental rats ICE-I model was made by injecting minim collagenase VII. Male \Vis tar rat, weighing 240 00g, were -divided into the four groups of normal, control, INN (middle dosage) and QKL. The treatment access was injection of abdominal cavity. All the data were recorded on the 1st, 3rd and 7th day. 2. 1 The effect of INN on nerve cell apoptosis of ICH experimental rats The terminal-deoxynucleotibyl transferase mediated nick end labeling (TUNEL) and image analysis were employed to detect the nerve cells apoptosis in the brain paraffin sections. Results showed that the positive cells of TUNEL stain increased notably in model group.
