Object:To investigate the mechanism of Liu-Shen-Wan Chan-Su and realgar in treating leukemia from the aspect of apoptosis.And to observe the different influences on liver of rat for the three durgs.Methods:The MTT assay was used to observe the growth inhibition on HL-60 cells of Liu-Shen-Wan, Chan-Su and realgar, the fluorescence and Wright-Giemsa dyeing assay were used to observe the morphologic changes of apoptosis, and the HE dyeing assay was used to observe the pathological changes of organs for rats, And the molecullar assay of RT-PCR was used to examine the mRNA level of gene bcl-2,c-myc and bax.Finally, the immunohistochemistry assay was used to examine the protein level of the three genes.Results:1.The water solution distilled from Liu-Shen-Wan and Chan_Su had obvious effects on inducing HL-60 apoptosis,while realgar had no such effect.The mixture of Peral, bezoar,musk and borneol had no effects of inducing apoptosis.2.Liu-Shen-Wan down-regulated the expression of c-myc and bcl-2 mRNA levels and up-regulated the expression of bax gene2083.Liu桽hen梂an reduced the toxic impair of the rats liver aroused by Chan桽u and reaglar.4.Liu桽hen條Van Induced the spleen and thymus of rats hyperplasia.Conclusion:Liu桽hen梂an is an effective prescription in treating leukemia. , and it may be through many approaches to achieve this effect ,and is worthy researched deeply.
