| Objective:To study the growth-inhibitory and apoptosis-induced effects of realgar nanometer suspension in human ovarian cell line SKOV3.Methods:Different concentration (6.25mg·L-1,12.5mg·L-1,25mg·L-1,50mg·L-1)of the realgar nanometer suspension used in SKOV3cells.After12h,24h,48h,72h, to observe the morphological changes of experimental group (with different concentration of realgar nanometer suspension treatment group) and control group (cells without drug)by optical microscope.To assay the growth suppression of the cells by Methyl thiazolyl tetrazolium(MTT).To detect the apoptosis percentage of the cells by the flow cytometry(FCM). To detect the expressions of Bcl-2/Bax by real time fluorescent quantitative polymerase chain reaction(RT-FQ-PCR).Results:After being treated with6.25mg·L-1,12.5mg·L-1,25mg·L-1,50mg·L-1realgar nanometer suspension for different time, the growth suppression of SKOV3cells appeared and there’s a time-dose relationship in a certain range. According to FCM, after being treated with50mg·L-1realgar nanometer suspension for48h, the apoptosis peak of SKOV3cells is obvious and the apoptosis rate of SKOV3cells is (25.83±1.42)%. According to RT-FQ-PCR, the expression of Bcl-2was down-regulated while the expression of Bax was up-regulated.Conclusions:Realgar nanometer suspension could inhibit the proliferation of SKOV3cells and induce its apoptosis.The apoptosis is related to the down-regulation of Bcl-2and up-regulation of Bax. |
