Section Ⅰ Improvement of the model of bromocriptine induced abortion rats and preliminary study on decidual apoptosis Objective: To improve the model of bromocriptine induced abortion rats and toexplore apoptosis of deciduae. Methods: Forty pregnant rats were randomized into four groups, 0.5 mg/kg·d,0.4 mg/kg·d, 0.3 mg/kg·d and 0.2 mg/kg·d of bromocriptine was injectedsubcutaneously during 6 to 8 days of gestation. All rats were sacrificed at the ninthday to observe the outcome of pregnancy. The dosage that cause the abortion rate ofover 70% would be accepted as the optimal dose. The deciduae of both normalpregnant rats and abortion rats were also scratched for TUNEL. Results: The abortion rate of 0.5 mg/kg·d, 0.4 mg/kg·d, 0.3 mg/kg·d and 0.2mg/kg·d of bromocriptine were 100%, 80%, 50% and 40%, respectively. So 0.4mg/kg·d was accepted as the optimal dosage. The ratio of TUNEL positive nucleolusto total nucleolus of normal pregnant group and the abortion group was (12.05±1.95)% and (41.36±2.87) %, (P<0.05). Conclusion: Injection of 0.4mg/kg·d of bromocriptine during 6 to 8 days ofgestation could make stable rat model of abortion. Deciduas apoptosis was observedin normal pregnant rats at the ninth day of gestation, but the apoptosis ratio ofabortion rats was higher than normal pregnant rats.Section Ⅱ The effect and mechanism of Chinese Herbs on deciduae apoptosis in bromocriptine induced abortion rats Objective: To study the deciduae apoptosis at different days of gestation andthe effect and mechanism of Chinese Herbs on deciduae apoptosis in bromocriptineinduced abortion rats. Methods: The pathological model of abortion rats was established as section Iand were randomized into three groups: Group Bro (n=10) was the abortion model.Group H (n=10) were treated with Chinese Herbs orally everyday for 9~12 daysstarting from the first day of pregnancy and Group P (n=10) were injected withprogesterone intramuscularly everyday for 3~6 days starting from 6th day ofgestation. Ten normal pregnant rats were set as control. All rats were killed at 9thand 12th day of gestation to observe the pregnant outcome. Deciduae apoptosis was detected byTUNEL and the expression of Caspase-8 and Caspase-3 mRNA was detected byusing RT-PCR. Caspase-3 activity was measured using Fluorometric Assay Kit.Expression of Bcl-2 and Bax proteins were detected by immunohistochemistry. Conclusion: The successful pregnancy rate of Chinese Herbs treatment was80% to 90% with no difference compared with Group N and Group p. Deciduaeapoptosis rate at 9th day of pregnancy was higher than that of 12th day (P<0.05).Expression of Caspase-8 and Caspase-3 mRNA and Caspase-3 activity and the ratioof Bax to Bcl-2 were also higher than that of 12th day (P<0.05). At the same time ofpregnancy, Deciduae apoptosis rate of Group Bro was significantly higher thanGroup N, Group P and Group H (P<0.05). Expression of Caspase-8, Caspase-3mRNA and Caspase-3 activity as well as the ratio of Bax to Bcl-2 protein were allhigh than Group N, Group P and Group H (P<0.05). Conclusion: Deciduae apoptosis rate at 9th day of pregnancy was higher thanthat of 12fth day. Excessive apoptosis was observed in bromocriptine inducedabortion rats while Chinese Herbs and progesterone could inhibit deciduaeapoptosis to prevent pregnant loss.
