Thrombus is main pathologic basis of many diseases, such as hypertension, myocardial infarction, cerebral ischemia and atherosis. Either induced by oxygen free radical or hypoxia, endothelial cells damage and its obstacle of haemostasis and thrombus dissolution, appear one of the main mechanisms of thrombus formation.Flavonoid compounds are low molecular weight ingredients in natural plant, and have diverse pharmacological actions with low side effect. Beside effects in preventing and curing cardio, cerebral vascular disease and breath disorder, it has activities of inhibiting bacterium, decreasing blood glucose, anti-oxidation, anti-radiation, anti-cancer and improving immunity. The flavonoid ingredients, such as rutin, quercetin, hypericin and vitexin, are main efficient components in Hawthorn leave, which reduce blood pressure and blood lipid, while improve cardiac output. However, the Hawthorn leave flavonoids in anti-thrombusis lacks the related prophase appraising.In order to develop the more efficient anti-thrombosis drug from traditional Chinese herb, expand its application and control its side-effect, the regulation actions of the extract of flavonoid-rich leave of Hawthorn (80% flavonoids, w/w) on endothelial cells were studied in this paper through flow cytometry mainly.The damage of endothelial cell is the basis of its function abnormality. In vivo, there are many complex factors inducing the endothelial damage, which include ROS and hypoxia. Thus mimicing factors inducing damage in vivo, in order to study the protective effect of the extract of flavonoid-rich leave of Hawthorn on HUVECs, we investigated its regulative actions (a) against HUVECs damage induced by ox-LDL; (b) on hydrogen peroxide in HUVEC_S; (c) on Ca~2+ and NO levels in HUVECs induced by hypoxia.In addition, as for appraising targets, the anti-thrombus formation factor- TM protein and the fibrin dissolutive factor - t-PA protein were examined in this study in vitro. Especially in the study of t-PA relative activity, we used an advancing model which eliminate the interfere in optical intensity measurement of traditional Chinese drug with color. This model is economical and veracious.Our study demonstrate that this extract can regulate endothelial cells in two aspects. On the one hand, by influencing the level of intracellular ion, increasing enzyme activity, scavenging ROS, inhibiting the expression of apoptosis protein and up-regulating anti-apoptosis protein expression, it protects the endothelial cells efficiently. On the other hand, it facilitates the expresssion and the relative activity of anti-thrombus factor. Moreover, this extract does not obviously impact the endothelial cells viability. In conclusion, the extract of flavonoid-rich leave of Hawthorn is worth being further studied and developed.
