Background and Objective:The clinical application of purification is widely increasing.In many this patients are often accompanied by haemorrhage tendency. But blood purification treatment must be dealed with anticoagulant. High risk of bleeding in patients with anticoagulant treatment has become a thorny issue. So,it is restricted by adverse reactions of anticoagulant in Blood purification to treatment with haemorrhage tendency patients.In the process of blood purification, the contact surface of blood dialyzer/filter is the key parts of the coagulation occurred. the surface properties of the filter membrane material determine the biological response of between the material with human blood and the organization, and determine the biocompatibility of the material as a whole and anticoagulant properties. Previous experiments of this group have been successfully using molecular Grafting and Crosslinking Argatroban small molecules grafted onto the polysulfone membrane surface, its anticoagulant modified, and confirmed that they have good biocompatibility.Such as McClungD contained the e-amino group of freedom of lysine (e-lysine) grafted onto biomaterial surfaces, confirmed a high degree of e-lysine combined with blood plasminogen (Plg), surface Plg activation rapidly changingplasminogen, which has the ability to dissolve clots.This study was to covalently bonded on the basis of preliminary studies, build the Argatroban-e-lysine modified membrane that has the properties anticoagulant and antithrombotic, and test its anticoagulant and antithrombotic performance,to explorean idea1anticoagulant antithrombotic modified filter membrane.Methods:1. Argatroban was covalently bonded on the surface of the polysulfone membrane by molecular graft crosslinking technology.2. The Argatroban modified membrane succinimide carbonate (DSC) the reaction of the surface modified membranes containing N-hydroxysuccinimide (NSC) groups, followed by substitution reaction and deprotection build Arg-e-lysine modified membrane3. XPS analysis detected membrane surface elements to track and analyze the process of modification, monitoring the process of modification.4. PS-Arg-Lys membrane recalcification test, and detection of activated partial thromboplastin (APTT), prothrombin time (PT), thrombin time (TT) and thrombolytic test in order to understand the anticoagulant and antithrombotic performance of the modifiedmembrane.Results:1. The result of the X-ray confirmed that it was successfully constructed anticoagulant antithrombotic modified polysulfone membrane by molecular cross-linking and covalent grafting method.2. PS-Arg-Lys membrane recalcificatic time and APTT compared with anticoagulant modified membrane and the unmodified membrane were significantly longer (P<0.05) was statistically significant, show that the e-lysine modified membrane with good anticoagulant properties.3. PS-Arg-Lys membrane TT than PS-Arg membrane and the unmodified membrane were extended (P<0.05) was statistically significant; PS-Arg membrane and the unmodified membrane TT time no difference (P>0.05), not statistically significant, indicating that the anticoagulant and antithrombotic properties of the PS-Arg-Lys membrane from the coagulation system and fibrinolytic system.4. PT of the three membranes have no difference (P>0.05), not statistically significant, indicating that the anticoagulant the antithrombotic modified membrane does not affect the extrinsic coagulation pathway.5. Thrombolytic test PS-Arg-Lys membrane than PS-Arg membrane absorbance peak low. PS-Arg-Lys membrane after reaching the peak, the absorbance gradually decreases and tends to baseline, indicating that anticoagulant anti-the suppository modified membrane has the ability to anticoagulant and antithrombotic.Conclusion: 1. The molecular grafting-cross-linking and covalent grafting method can be successfully modified the PS-Arg-Lys membrane2. PS-Arg-Lys membrane has a good anticoagulant properties3. PS-Arg-Lys membrane has a certain antithrombotic properties...
