Background Coronary heart disease (CHD) is one of the hazardous diseases whichare harmful to people and it is the leading cause of death in the world, causing almost15,000,000 deaths annually and its annual treatment cost is over 50 billions dollars. In ourcountry, it causes almost 1,500,000 deaths annually, and its annual treatment is over 20billions Yuan in RMB. As a result,it brings a burden to the nation's economic expense.Angina cordis is the main symptom of CHD, so studies on CHD are of great importance. Inthe present, western medicine is widely used to treat CHD,but side effects and recurrenceoften show up. However, treatment with Chinese medicine is superior. Considering itssensitivity in old people, we first propose the renal deficiency and blood stasis type CHD,and devise San Baoxin capsule to cure CHD. To estimate the effect of San Baoxin capsuleon CHD, we need a convenient animal model. In the present, coronary artery legation is themost popular method to make CHD in rat. In the model, we could observe the changes ofischemic cardiac muscle as well as estimate drug effects quantitatively. But the model iscomplex to make and the survival rate of animals is low. Therefore, we must improve thepresent animal model before estimate the effect of San Baoxin capsule on CHD. Objective To simplify the CHD model in rat and to decrease the death rate ofanimals; to estimate the effect of San Baoxin capsule on rat CHD model, rat reperfusionmodel, dog CHD model, dog hemodynamics, dog myocardial consumption of oxygen,rabbit platelet aggregation and rat blood apparent viscosity. Methods To improve the CHD model made by coronary artery legation: ①Etherization and chloral hydrate were used simultaneously for anesthesia and oral tracheacannula was performed.② Breathing machine was omitted to simplify the CHD model. To estimate the effect of San Baoxin capsule on CHD, haemodynamics and plateletaggregation etc:① The effect of San Baoxin capsule on CHD model in dog: San Baoxincapsule was administered through dodecadactylon, the scope and degree of myocardialischemia were measured through epicardial electrogram and the level of blood endothelin(ET), thromboxan B2 (TXB2) and 6-Keto-PGF1 were determined by radioimmunity during αthe three hours after administration, three hours after administration, the scope ofmyocardial infarction were measured by staining method with nitro blue tetrazolium stain(N-BT stain), the above indexes were compared between San Baoxin groups and theCHD model group. ②The effect of San Baoxin capsule on haemodynamics andmyocardial oxygen consumption in dog: San Baoxin capsule was administered throughdodecadactylon, the coronary blood flow, cardiac output, levo-intraventricular pressure, themaximal upstroke velocity of levo-intraventricular pressure, arterial and venous bloodpressure, electrocardiogram and myocardial oxygen consumption were measured bypolygraph during the three hours after administration, the above indexes were comparedbetween San Baoxin groups and control group. ③ The effect of San Baoxin capsule onmyocardial ischemia-reperfusion model in rat : three hours after intragastric administration,blood was obtained from abdominal aorta, the level of serum lactate dehydrogenase (LDH)and creatine kinasea (CK) were determined by radioimmunity, the infarct size wasmeasured by staining method with N-BT stain, the above indexes were compared betweenSan Baoxin groups and CHD model group. ④The effect of San Baoxin capsule on plateletaggregation in rabbit : blood was obtained from auri-artery, packing fraction of platelet wasmeasured before and after intragastric administration of San Baoxin capsule. The differencebetween the packing fraction before and after intragastric administration was comparedbetween San Baoxin groups and control group. ⑤The effect of San Baoxin capsule onblood apparent viscosity in rat: intragastric administration was performed once a day forseven days.
