Xiongzhidilong Soup Migraine Model Animal Analgesic Sedative Effect And Mechanism

Objective:To study the analgesic effect and the sedation of Xiong Zhi Di Long Tang on migraine model animal,and investigate the mechanism of Xiong Zhi Di Long Tang in treating migraine from this aspects as follows: neurobiochemical metabolic disorder and nociceptive pain information transmitting in head.Method:The annagesis effect of Xiong Zhi Di Long Tang was study with hot plate inducing algesia and acetic acid inducing writhing response.The sedation of Xiong Zhi Di Long Tang on nitroglycerin inducing migraine model was study with tail-suspended test and the analytical system of animal behavior-spontaneous movement.The effect of Xiong Zhi Di Long Tang on neurobiochemical metabolic disorder in nitroglycerin-inducing-migraine-model was study through determining the contents of CGRP,β-EP,NO and NOS in plasma and brainstem.The effect of Xiong Zhi Di Long Tang on algesic transmission in nitroglycerin induced migraine model was study through analysis neuronal emotional state and coexistence express of CGRP,β-EP,NOS and 5-HT which located at spinal nucleus of trigeminal nerve and periaqueductal gray matter.Results:Xiong Zhi Di Long Tang can prolong the pain threshold of mice which were induced algesia by hot,weaken writhing response of mice which were induced by acetic acid.This effect is more significant in large doses group and middle doses group. Xiong Zhi Di Long Tang can prolong tail-suspended quiescent time of mice which were induced migraine by nitroglycerin,shorten active path and active time of mice which were induced migraine by nitroglycerin. This effect is more significant in middle doses group. Xiong Zhi Di Long Tang can cut down the heightened contents of CGRP,NO and NOS in plasma and brainstem of rats which were induced migraine by nitroglycerin,increase the decreased contents ofβ-EP in plasma and brainstem of rats which were induced migraine by nitroglycerin.This effect can maintain a equilibrium level all the time. Xiong Zhi Di Long Tang can inhibit concurrent expression of c-fos and NOS,c-fos and CGRP in spinal nucleus of trigeminal nerve and periaqueductal gray matter of rats which were induced migraine by nitroglycerin,increase expression of 5-HT,β-EP in spinal nucleus of trigeminal nerve and periaqueductal gray matter of rats which were induced migraine by nitroglycerin.This effect inhibit nociceptive pain information transmitting.Conclusion: Xiong Zhi Di Long Tang both have central analgesis effect and peripheral analgesis effect.
