Guan Mu Tong Compatibility In Longdan, Renal Toxicity Studies

Long Dan Xie Gan Decoction (LDXGD,《Yi Fang Ji Jie》)consisted of ten herbs, Gentiana scabra Bunge. Scutellaria baicalensis Georgi. Gardenia jasminoides Ellis. Alisma arientalis (Sam.) Juzep. Aristololochia manshuriensis Kom. Plantago asiatica.,Angelica sinensis. Rehmannia glutinosa Libosch. Bupleurum chinensis DC. Glycyrrhizae uralensis. It can purge hepatic-cholic fire and expel heat and dampenss there,on the other hand,it avoid the hepatic hurt and the consumpion of Yin. It is used in clinic as decoction or pill wind–ranging. However ,there were some renal damage records caused by Long Dan Xie Gan Pill (LDXGP)recently,and they aroused public attraction, accordingly,the nephrotoxicity of LDXGP/ LDXGD has become the hot spot caused world-wide attention.According to National Pharmacopeia (2000 yr.), LDXGP or LDXGD can contain Aristololochia manshuriensis Kom(AMK). AMK belongs to Aristolochiaceae , and affects renal damage ,therefore, it was concluded that LDXGP or LDXGD has nephrotoxicity. Nevertheless,it is not reasonable that estimate the toxicity or side effect of the traditional Chinese medicine and prescription only according to one ingredient of herb or one herb in prescription. This method is not consistent with clinic experience of traditional Chinese physician or empirical study at present.In this reseach ,we want to explore the nephrotoxicity of LDXGD and find out the relationship of the compatibility of AMK in different dosages with the prescription ,and illuminate the possible mechanism of the nephrotoxicity of LDXGD. We try to provide more safe guidance in clinic and set an example for the research about the toxicity or side effect of the traditional Chinese medicine and prescription.This dissertation contains the documents review and the experiment. The clinical and experimental research of Aristololochia manshuriensis Kom is summarized in the document part. The relationship between dosage ,administer-time , compatibilition with the nephrotoxicity ,and the nephrotoxicity mechanism are discussed in the experimental part.
