Objective To examine the levels of plasma homocysteine and the expression of NF-κB in aorta from methionine -fed rabbits and the effects of Guanxinkang on atherosclerotic lesion and NF-κB.T0 investigate the antiatherogenic effect of Guanxinkang .Methods Twenty-four rabbits were randomly divided into three groups. The control group were fed on common food 100g a day,the Hcy model were fed on high-methionine food which composed of common food containing 1% methionine 100g a day,while the Guanxinkang group with high-methionine food 100g/d+Guanxinkang powder 1.45g/kg/d. Blood samples were collected before (time 0) and after 4 and 8 weeks of experimental diets for measurement of serum triglycerides, total cholesterol (TC), and LDL, HDL cholesterol (LDL-C, HDL-C), and plasma hcy. At the end of 8th weeks, the animals were put to death to establish pathological model of aorta for light microscopy to observe the vascular atherosclerotic lesion and its degree. The NF-κB expression in aortas was using ABC (Avidin-Biotin Complex) and DAB area ratio of the positive expressing cells of NF-κB (NF-κB%)was analyzed with Image-Pro Plus Image Analysis System and The relative expressing level of the mRNA expression of NF-κB was defined by optical density ratio of NF-κB to GAPDH respectively (NF-κB/GAPDH) by RT-PCR.Results(1) The level of plasma Hcy and serum TC, TG, LDL-C, and the ratios LDL-C/HDL-C and TC/HDL-C in the model group significantly higher than the control group(p<0.05), and markedly expression of NF-κB mRNA in aorta can be detectived by Immunohistochemistry. The endothelia cell hyperlasia on intima can be observed ,tunica media was infiltrated by the dot and local lipid . (2) Guanxinkang remarkably decreases the lipid in serum and plasma Hcy , inhibits NF-κB mRNA expression in aorta(P<0.05),also can attenuate aortic intimal proliferation.Conclusions:(1)The model of hyperhomocysteinemia(HHcy) can be established after high-methionine diet for 8 week. The HHcy can induce atherosclerosis in rabbits. (2) Guanxinkang markedly decreases the lipid in serum and HHcy, also inhibits NF-κB expression. Guanxinkang can prevent and theapy the HHcy, diminish the artery damage.
