Object:This experiment, having the energy metabolism of mitochondria as a main line,researched the change of mitochondria codes gene expression in endotoxemia,the variety of relevant enzyme activity in oxidative phosphorylation, mitochondria Oxidative injury and mitochondria membrane potential.It would discuss the possible mechanism of Jie Du Hua Yu Fang improving the mitochondria energy metabolism obstacle in endotoxemia.Method: 56 Wistar rats were randomly divided into 7 groups, including the normal group and 6 ,12, 24 h groups after endotoxin and Jie Du Hua Yu Fang 6 , 12, 24 h groups.Each group had 8 rats. We druw liver organization and observed through electronic microscope and optical microscope, examined the level of ALT, AST through automatic bio-chemical analytical instrument, measured the level of TNF-α through the method of ELISA, measured the content of ATP,ADP,AMP in the mitochondria of liver cell through the method of high pressure liquid chromatography, examined the content of MDA and the activity of SDH, Na+K+-ATPase, Ca2+Mg2+-ATPase, SOD, GSH-PX, COX in the mitochondria of liver cell through spectrophotometry instrument,measured hepatocyte mitochondrial tran- smembrane potential( Δψm) through the flow type cell instrument, examined the changes of the expression of liver mitochondria mtDNA, COX Ⅰ, COX Ⅱ, COX Ⅲ, ATPase6, ATPase8mRNA through the method of RT- PCR.Result: 1. The level of ALT, AST went up gradually in 6 , 12, 24 hours groups after endotoxin, comparing with the normal group,which had Statistics Meaning(P<0.05 or P<0.01). The level of ALT, AST in Jie Du Hua Yu Fang 12h,24h groups was lower than it in the groups of endotoxin(P<0.01). The optical microscope results manifested: after endotoxin ,Hepatic sinusoid extended, hepatocyte was mutation and dead, the inflammation cell appeared in portal area, and the pathological changes of the liver histology had obviously improved in Jie Du Hua Yu Fang groups. The electronic microscope results manifested: after endotoxin,Hepatic mitochondria swelled, mitochondrial cristae absented and was ballooning degeneration, and the super microstructural pathological changes of the liver cell had obviously improved in Jie Du Hua Yu Fang groups.2.After endotoxin,the content of ATP in hepatic mitochondria descended gradually in 6, 12, 24 hours groups, comparing with the normal group, which had Statistics Meaning (P<0.01). The content of ATP in the group of 24h after endotox, which was only 23% of the normal group, was the lowest; The content of ADP descended too, which had Statistics Meaning (P<0.01); The content of AMP increased after six hours, descended after twelve hours, and arrived at the lowest point after twenty-four hours, comparing with the normal group,which had Statistics Meaning (P<0.01); the content of hepatic mitochondria EC descended gradually, comparing with the normal groups, which had Statistics Meaning(P<0.01). The group of 24h in Jie Du Hua Yu Fang compared with the group of 24h after endotoxin ,the content of ATP and EC all increased, comparing with each other, which all had...
