Part OneEffects of Celecoxib on Expressions of Cyclooxygenaseand Nuclear Factor-kappa B in Cultured HumanUmbilical Blood MonocyteBackground: It's well known that atherosclerosis(AS) is a inflammatory disease, but no optimal strategy has been found aimed at it directly. Cyclooxygenase(COX)-2 inhibitor is a potential agent which can possibly retard AS owing to the close relationship between COX-2 and AS as well as inflammation. Monocyte/Macrophage is the most important cell type representing inflammatory process during AS. To study further whether COX-2 inhibitor can retard AS through its anti-inflammatory effect, it is an important groundwork to study the effect of COX-2 inhibitor on Monocyte/Macrophage first.Objective: To study the effect of Celecoxib, a COX-2-specific inhibitor, on expressions of COX-1, COX-2 and nuclear factor-kappa B (NF-kB) in Interleukin-1β (IL-1β) stimulated human umbilical blood monocyte (HUBMC) cultured in vitro.Methods: HUBMC were isolated and cultured by Ficoll density gradient centrifugation and adhering method, then divided into groups as follows: Blank Control group, IL-1β + DMSO group, IL-1β+ Celecoxib group, IL-1β + Aspirin group. Cells were pre-incubated with Apirin in 1% DMSO(0.05mM, 0.1mM, 0.5mM, 1.0mM), Celecoxib in 0.1% DMSO(1.0μM, 2.5μlM, 5μM, 10μM), or DMSO(1% for Apirin group and 0.1% in v/v for Celecoxib group as control) for 1 hour respectively, then stimulated with IL-1(3(10ng/ml) and collected 16 hours after adding...
