Effects Of Cyclooxygenase-2 Inhibitor Celecoxib On The Expression Of VEGF In Human Cholangiocarcinoma QBC939 Cell Lines

Purpose:This study aims to observe the effects of the selective cyclooxygenase-2(cox-2) inhibitor,celecoxib,on the expression of vascular endothelial growth factor(VEGF)in human cholangiocarcinoma QBC939 cell lines,thereby probe into the mechanisms of cox-2 and the selective cyclooxygenase-2 inhibitor in in human cholangiocarcinoma QBC939 cell lines.Methods:The time-effect relationship and concentration-effect relationship between Celecoxib and the expression of VEGF in human cholangiocarcinoma QBC939 cell lines were studied by using reverse transcription polyrnerase chain reaction(RT-PCR)and enzyme-linked immunoadsorbent assay(ELISA).Results:1.The expression of VEGF mRNA in QBC939 cells were significantly inhibited by celecoxib in a certain dose(20～80 umol/L),compared with control group(P＜0.05).2.The concentration of VEGF in the supernatant were evidently decreased in dose-dependent manner in celecoxib(20～80 umol/L)treated groups,compared with control group(P＜0.05).3.The expression of VEGF mRNA in QBC939 cells were markedly reduced in 40 umol/L celecoxib treated groups(12～48h),compared with control group(P＜0.05).4.40 umol/L celecoxib treating for a certain time(12～48h)prominently decreased the concentration of VEGF in the supernatant,compared with control group(P＜0.05).Conclusions:The human cholangiocarcinoma QBC939 cell lines can express VEGF.COX-2 may play an important role on the expression of VEGF in human cholangiocarcinoma QBC939 cell lines,while the selective COX-2 inhibitor,celecoxib,can inhibit the expression of VEGF in a certain degree.
