Objective: To detect the expression of cyclooxygenase 2(COX2)and and nucler factor-kappa B(NF-κB) protein and m RNA in gastrointestinal stromal tumors(GISTs),investigate the relationship between COX2,NF-κB and clinicopathologic features and the role of COX2 and NF-κB in the prognosis of GISTs. Methods: Immunohistochemical En Vision technique was used to detect the expression of COX2 and NF-κB protein and in situ hybridization(ISH)method was used to detect COX2 and NF-κB m RNA expression in GISTs. The Kaplan-Merier method was used to analyze the follow-up data of the cases of GISTs.Results:In 119 cases of GISTs:(1)97 cases positively expressed COX2 protein, the positive expression rate was 81.5%.The difference of expression of COX2 protein in different risk classification was statistically significant(P<0.05),but in different gender,age,location, histological type was not statistically significant(P>0.05).( 2) 98 cases positively expressed NF-κB protein, the positive expression rate was 82.4%.The difference of expression of NF-κB protein in different risk classification was statistically significant(P<0.05),but in different gender,age,location, histological type was not statistically significant(P>0.05).(3) By selecting 78 cases in all of the 119 cases of GISTs for ISH:60 cases positively expressed COX2 m RNA, the positive expression rate was 76.9%. 61 cases positively expressed NF-κB m RNA, the positive expression rate was 78.2%.The difference of expression of COX2 and NF-κB m RNA in different risk classification was statistically significant(P<0.05),but in different gender,age,location, histological type was not statistically significant(P>0.05).(4)By rank correlation analysis of data, the expression of COX2 and NF-κB protein and GISTs risk classification had positively correlation(P<0.05); COX2 and NF-κB protein had correlation(r=0.461,P < 0.000),as well as COX2 and NF-κB m RNA(r=0.227,P < 0.000) in GISTs.(5)65 patients were followed up,the follow-up rate was 54.6%.By Kaplan-Meier survival analysis of GISTs with follow-up data, the difference between the different expression of COX2 and NF-κB protein was significant(P<0.05), but had no difference in terms of the survival rate(P>0.05). There was higher expression of COX2 and NF-κB protein in higher rate group of metastasis. Conclusions:1. The positive expression of COX2 and NF-κB protein and m RNA exists generally,and is associated with the risk classification of GISTs, COX2 and NF-κB may promote the ocurrence and development of GISTs. 2. Protein and m RNA expression of COX2 and NF-κB are positive correlated,the synergy of both could promote the development of GISTs. 3. COX2 and NF-κB protein expression is closely related to the prognosis of the GISTs,can be used in the prognostic evaluation of GISTs.
