Interleukin-1 (IL-1) is an important cytokine, which has many biological activities. It works through its high affinity interaction with IL-1 receptor, which is present on a wide variety of different cell types. There are two types of IL-1 receptors: type I rceptor(IL-1R I ) and type II receptor(IL-1R II). Soluble forms of IL-1R can be produced through protelytic cleavage of the extracellular portion of the two types of receptors.Tumor necrosis factor (TNF) is an other kind of important pre-flammatory cytokines, there are also two types of TNFR: TNFR I and TNFR II. Soluble forms of TNFR were generated by protelytic cleavage extracelluar protion to downmodulate membrane-bound receptors of the two type receptors: sTNFR I and sTNFR II.There are two sections in the project.In the first section, the metabolites of streptomyces 139 have been well studied. From the IL-1R antagonist screening model, a kind of inhibitor of IL-1R, 139A was obtained. Its extraction method was refined. Through chemical analysis, chromatography, spectral analysis, 139A was elucidated as a kind of polysacchride and its structure was deduced.In the second section, the metabolites of fungus 1487 has been well studied. It was obtained from the TNFR antagonist screening model targeting the recombinant human sTNFR expressed from recombinant streptomyces lividans(pIJ459-mel/ sTNFR).
