Objectives:Recent years,incidence rate of the crebrovascular disease ecurrent condition has a sharp increasing, and its disability rate and case fatality rate appears much higher. Atherosclerosis is a fundamental symptom of crebrovascular disease, and metabolic disorder of blood fat would resulted of artherosclerosis.This theory has been revealed in many research reports in the world; Anormal condition of blood fat is deeply related with crebrovascular disease. Hyperlipidemia is a basic athological element of weighted artherosclerosis, and also is the major criteria of causing artherosclerosis condition getting worse.Hyperlipidemia is the one third portion of recurrent of cerebral apoplexy in the incidence rate.Therefore, to reduce blood fat is a significant point in decreasing recurrent of crebrovascular disease.In clinical practices, most of Cerebrovascular disease patients have hyperlipidemia, so when these kind of patients havecerebral apoplexy, their condition of cerebral ischemia always get worse because of their metabolic disorder of blood fat , even worse than the condition of cerebral ischemia only.To effectively regulate the level of blood-lipid can prevent the occurrence of cerebrovascular disease. therefore, it is one of the most important subject of clinical and experimental research to find the effective preventing and treating methods for cerebrova- scular disease.Methods: (1) The rats were fed on high fat standard forage to make hyperlipemia. Then,the model of hyperlipemia with cerebral ischemia was established by Middle CerebralArtery Trumbosis(MCAT). Rats divided into eight gropes in random. Preoperative intervention and the whole range after cerebral ischemia with electropuncture treatment.(2) Use biochemical method to observe the change of blood-lipid and the influence of acupuncture after cerebral ischemia.(3) Use neurobehavioral index to compare the symptoms of pretreatment and post-treatment, and thereby work out the statistical estimation. Use morphologic method to observe the pathological change condition of the ischemic side and the hippocampus area with the acupuncture effects.(4) Use immunohistochemical method to assay the GFAP,HSP70,S-100βand vimentin expression of astrocyte in the ischemic side and the hippocampus area before and after acupuncture. (5) At the end of the experiment, use ABC-ELISA method to detect the level of NGF,BDNF and bFGF in the brain of the rats.Results:1.High fat standard forage fed hyperlipemia model rats is very ideal in laboratory research. Then,the model of hyperlipemia with cerebral ischemia was established by Middle Cerebral Artery Trumbosis(MCAT).The four blood-lipid indexs can be used as the examming method to analysis hyperlipemia.2.Positive neurobehavioral reaction can be found in one to two hours after ischemia. The neurobehavioral score of all ischemic model groups significantly higher than normal and high blood lipid groups and also higher than all acupuncture groups. The study shows that acupuncture therapy can reduce the neurobehavioral score and improve neural founction.3.Acupuncture therapy can regulate the level of blood-lipid.4.As compare with the normal group by using the rat, which was the condition of the hippocampus area and the ischemic side of the simple hyperlipemia, it was appeared that the the mean number of immunopositive cell,area density and optical ensity of the GFAP,HSP70,S-100βand vimentin of the hyperlipemia model group was reduced. To compare with the after 7 days acupuncture treatment, hyperlipemia treatment group, the mean number of immunopositive cell,area density and optical ensity of the GFAP,HSP70,S-100βand vimentin is increased.After the damaged of the ischemia occurred, it was significant increased on the the mean number of immunopositive cell,area density and optical ensity of the GFAP,HSP70,S-100βand vimentin of the hippocampus area and the ischemic side of the rat. To compare with cerebral ischemia model group after 7 days acupuncture treatment, the mean number of immunopositive cell,area density and optical ensity of the GFAP,HSP70,S-100βand vimentin of cerebral ischemia treatment group was increased.After hyperlipemia rat brought to hyperlipemia with cerebral ischemia model group, to compare with cerebral ischemia model group, the the mean number of immunopositive cell,area density and optical ensity of the GFAP,HSP70,S-100βand vimentin of the hippocampus area and the ischemic side of hyperlipemia with cerebral ischemia model group was decreased. To compare with hyperlipemia with cerebral ischemia model group, the mean number of immunopositive cell,area density and optical ensity of GFAP,HSP70,S-100βand vimentin of hyperlipemia with cerebral ischemiaⅠ-treatment group and hyperlipemia with cerebral ischemiaⅡ-treatment group was sharp increased, p<0.01; The comparison with hyperlipemia with cerebral ischemiaⅡ-treatment group, each items of GFAP,HSP70,S-100βand vimentin of hyperlipemia with cerebral ischemiaⅠ-treatment group had marked increased, p<0.01。5.The contents of NGF,BDNF and bFGF: cerebral ischemia treatment group> cerebral ischemia model group> normal comparison group . normal comparison group> hyperlipemia treatment group > hyperlipemia model group.The contents of NGF,BDNF and bFGF in the hyperlipemia model group and hyperlipemia with cerebral ischemia model group were markedly lower than that in the normal comparison group (P<0.01).As compared with hyperlipemia with cerebral ischemia model group, the contents of NGF and bFGF was significantly higher in the hyperlipemia with cerebral ischemiaⅠ-treatment group and in the hyperlipemia with cerebral ischemiaⅡ-treatment group (P<0.01).As compared with hyperlipemia with cerebral ischemia model group, the contents of BDNF was significantly higher in the hyperlipemia with cerebral ischemiaⅠ-treatment group (P<0.01). There was no difference between hyperlipemia with cerebral ischemia model group and hyperlipemia with cerebral ischemiaⅡ-treatment group (P>0.05),but the contents of BDNF in hyperlipemia with cerebral ischemiaⅡ-treatment group was higher.The contents of NGF and BDNF in the hyperlipemia with cerebral ischemiaⅠ-treatment group was significantly higher than that in the hyperlipemia with cerebral ischemiaⅡ-treatment group (P<0.01). The contents of bFGF in hyperlipemia with cerebral ischemiaⅠ-treatment group was higher than that in thehyperlipemia with cerebral ischemiaⅡ-treatment group.Conclusion:1.The rats were fed on high fat standard forage to make hyperlipemia.Then,the model of hyperlipemia with cerebral ischemia was established by Middle CerebralArtery Tru- mbosis(MCAT). To build up hyperlipemia with cerebral ischemia model group succes- sfully would provide the various empirical study on the influence of the acupuncture f- or the hyperlipemia with cerebral ischemia rat at the astrocyte in the future, and also c- ould provide the accurate animal model on the study of pathogenesis of hyperlipemia with cerebral ischemia.2.Hyperlipemia can worsen the level of ischemic damage.3.Acupuncture therapy can regulate the level of blood-lipid.4.The high-level of GFAP,HSP70,S-100β,vimentin expression of astrocyte and the high-level of NGF,BDNF and bFGF expression in the brain may be related to the me- chanism of neuronic injury and repair. The electro- acupuncture may have protective e- ffects on hyperlipemia with cerebral ischemia. Time of interventional therapy is very i- mportant when using the electro- acupuncture.The electro- acupuncture may be used to treat hyperlipemia with cerebral ischemia at the early stage. Electro- acupuncture can up-regulate the long-term contents of GFAP,HSP70,S-100β,vimentin of astrocyte and can up-regulate the long-term contents of NGF,BDNF and bFGF in the brain, wh- ich may contribute to the effect of electro- acupuncture in improving hyperlipemia wi- th cerebral ischemia.
