Objective:To primarily learn of the expression of OY-TES-1 in kinds of tissue and the significance of OY-TES-1 antibody in serum, analysis of the relation of OY-TES-1 and tumor' occurrence and development,the relation of antibody against OY-TES-1 and tumor or other diseases,to explore the opposibility of applying OY-TES-1 in tumor immunotherapy.Menthods:We constructed the prokaryotic expression recombinant plasmid of OY-TES-1 and expressed and purifed OY-TES-1 fusion protein by the technique of genetic engineering.OY-TES-1 was analysed by Bioinformatics.The New Zealand white rabbit was immunized with the purified fusion protein to prepare polyclonal antiserum.We learnt the gene expression characteristic of OY-TES-1 in mRNA and protein lever by real time PCR and immunohistochemistry staining.The antibody against OY-TES-1 in nomal individuals sera and different sera of the patients with malignant tumor and other diseases such as hepatocirrhosis and needle-blocking were detected using purified OY-TES-1 as the antigen by ELISA,then we made a preclinic analysis on OY-TES-1.Result:The fusion protein MBP/OY-TES-1 was successfully acquired and sequensed to the target one by MS/MS.Although OY-TES-1 mRNA ubiquitously expressed in nomal tissue,the highest one was in testis that was over 90 times high in other nomal tissue.The expression level of OY-TES-1 in human HCC,meningioma and glioma was higher than adjacent carcinoma tissues,and no relationship was found between the expression of OY-TES-1 genes and clinicopathological parameters.We found no OY-TES-1 protein in many nomal tissue or cell except the renal tubule of one kidney,and no positive reaction was found in testis,colon,lever,skeletal muscle,stomac,lens,and leukocyte.25%(1/4)hepatocirrhosis and 50%(4/8)testis of needle-blocking were positive while other non-malignant tissue such as prostatitis and chronic hepatitis were negative.The expression rate of OY-TES-1 protein in HCC,gastric carcinoma,and colonma tissue was 40%(4/10),33.3%(1/3)and 25%(4/16),respecively,but 7 spacimens of lung cancer showed negative.The positive rate of OY-TES-1 antibody was 14.9%in the serum of tumor patient(including esophagus carcinoma 30%and HCC 20%)and 70.8%in needle-blocking while no OY-TES-1 sera reactivity was found in 76 nomal individuals or 17 hepatocirrhosis.Relation of OY-TES-1 antibody and needle -blocking showed siginificant difference.Conclusion:OY-TES-1 is a tumor associated antigen which is restrictly expressed in normal tissue and selectively highexpression in magnent tissue,It is a potential target for tumor immunotherapy.The relationship between OY-TES-1 and needle-blocking need further study.
