As a global public health problem,malignant tumor is a serious threat to human health.And the incidence and mortality rate showed a rising trend year by year.According to the latest data released by the World Health Organization,in 2012 the world's 14 million new cancer patients,of which more than 20% in china.As one of the common malignant tumors of digestive tract,the incidence and mortality of colon cancer is increasing year by year.Because of the complex mechanism of tumor development,it is very difficult and challenging to find effective treatment methods.In recent years,with the deepening of the research on tumor immunotherapy,immunotherapy has become the fourth treatment methods after surgery,radiotherapy and chemotherapy.This therapy can stimulate the specific immune response by enhancing the immunogenicity of tumor antigens,and then attack the tumor cells.Melanoma associated antigen(MAGE)is such a group of tumor specific antigen.The research group led by tutor has been devoted to the research of MAGE-A family in recent years.In this study,we proposed from the organization perspective,through the immunohistochemical staining method of MAGE-AS(including MAGE-A1,-A2,-A3,-A4,-A6,-A10 and-A12)expression in cancer tissues and corresponding adjacent colorectal tissues,and analyze the relationship between its expression and clinical pathology in patients.Colon cancer index and prognosis,and provide a new theoretical basis for the diagnosis and treatment of colon cancer.The main research contents and results are as follows:Objective: To investigate the expression of melanoma associated antigen MAGE-As(including MAGE-A1,MAGE-A2,MAGE-A3,MAGE-A4,MAGE-A6,MAGE-A10 and MAGE-A12)in colon cancer and its adjacent tissues.Methods: MAGE-As was detected by immunohistochemical method in 60 cases of colon cancer tissues and cancer adjacent tissues(including MAGE-A1,MAGE-A2,MAGE-A3,MAGE-A4,MAGE-A6,MAGE-A10 and MAGE-A12)protein expression and analyze its relationship with clinical biological behaviors and with the prognosis of patients.Result:1.MAGE-As(MAGE-A1,MAGE-A2,MAGE-A4,MAGE-A3,MAGE-A6,MAGE-A10 and MAGE-A12)protein was expressed in testis and colon cancer tissue.Positive expression of spermatogonia and spermatocytes in testis.2.MAGE-As(MAGE-A1,MAGE-A3,MAGE-A4,MAGE-A6,MAGE-A2,MAGE-A10 and MAGE-A12)protein was mainly localized in the cytoplasm of the cancer tissue,localized in the nucleus.The expression of MAGE-As(including MAGE-A1,-A2,-A3,-A4,-A6,-A10 and-A12)was not found in the adjacent tissues of 60 cases.3.the positive rate of MAGE-As(MAGE-A1,MAGE-A2,MAGE-A3,MAGE-A4,MAGE-A6,MAGE-A10 and MAGE-A12)in colon cancer was 56.7%(34/60)(= =0.087,P = 5).In colon cancer,MAGE-As(including MAGE-A1,MAGE-A2,MAGE-A3,MAGE-A4,MAGE-A6,MAGE-A10 and MAGE-A12)protein expression and age,gender,clinical results of patients with colon cancer staging(P>0.05),but no correlation with histological type and lymph node metastasis(P < 0.05)showed a positive correlation.Conclusion: MAGE-As may be a tumor associated antigen of colon cancer,and may be used as a the potential biomarker for prognosis colon cancer.
