Rhizoma Polygoni Cuspidati has the effect of promoting blood flow and transforming stasis, which can be used to treat diabetes and diabetic nephropathy. Anthraquinone, one of the essential components of Rhizoma Polygoni Cuspidati, has wide biological activities. The research is to study the early intervention of Anthraquinone of Rhizoma Polygoni Cuspidati on diabetic nephropathy.ObjectiveTo study the early intervention of anthraquinone of Rhizoma Polygoni Cuspidati on diabetic nephropathy and reveal its mechanism. MethodsFour mouse models were built, i.e. hyperglycaemia model caused by adrenaline and alloxan, diabetic nephropathy model and syndrome of blood stasis of the model caused by streptozotocin. The mice were randomly assigned into blank group, model group, control group, treatment groups (large, middle and low dose of anthraquinone groups). dimethyl biguanide and captopril were used to be the positive control drugs to research the hypoglycemic effect and kidney protection effect of anthraquinone of Rhizoma Polygoni Cuspidati. Serum levels of BG, BUN, Chol, H DL-D, TC, LDL-D,and AGEs, RAGE, TGF-β1, Smads in renal were detected. Light microscope was used to observe the pathological change of the kidney.Results1 The effect on hyperglycaemia model caused by adrenaline①Dimethyl biguanide can significantly decrease serum glucose level of 30 min and 60min after taking medicine(P﹤0.01).Different dosage of anthraquinone of Rhizoma Polygoni Cuspidati all can significantly decrease serum glucose level(P﹤0.05).②There is no significant difference between different dosage of anthraquinone in serum glucose level of 30min and 60min after taking medicine(P﹥0.05). And there is significant difference between control group and treatment group in serum glucose level(P﹤0.01).③The serum glucose lowering effect of dimethyl biguanide is superior to an -thraquinone of Rhizoma Polygoni Cuspidati. Middle dosage of anthraquinone can significantly increase the hepatic glycogen level.2 The effect on hyperglycaemia model caused by alloxan①The serum glucose level of the model group increases and hepatic glycogen level decreases, which indicates the success of model building.②All the dosages of anthraquinone and dimethyl biguanide can significantly decrease serum glucose level(P﹤0.01).③Middle dosage of anthraquinone can significantly increase the hepatic glycogen level(P﹤0.01).3 Kidney protection effect on diabetic nephropathy model①Anthraquinone of Rhizoma Polygoni Cuspidati can improve some symptoms like polydipsia,polyphagia and body weight loss of diabetic nephropathy model.②The treatment groups all can significantly decrease the kidney index (P<0.05).③High dosage and middle dosage of anthraquinone can significantly lower blood urea nitrogen level and serum creatinine level(p﹤0.05).④All the dosages of anthraquinone can lower serum glucose level after 4 weeks of medication.⑤High dosage and middle dosage of anthraquinone can not significantly decrease CHOL level and increase HDL level (p﹥0.05)and high dosage of anthraquinone can lower TG level(p﹤0.05). Middle dosage of anthraquinone can significantly lower TG and LDL leve(lp﹤0.01). It seems that low dosage of anthraquinone and captopril can decrease CHOL, TG and LDL level and increase HDL level, but there is no statistically significan(tp﹥0.05).4 Effect on AGEs,RAGE of diabetic nephropathy modelIn the model group, AGEs level and the expression of RAGE increase significantly. High dosage and middle dosage of anthraquinone and captopril can significantly decrease their level(p﹤0.05).5 Effect on TGF-β1,SMAD3 of diabetic nephropathy modelIn the model group, the expression of TGF-β1 and SMAD3 increase significantly while middle dosage of anthraquinone and captopril can significantly decrease the expression SMAD3(p﹤0.05).6 Kidney protection effect on diabetic nephropathy model①Anthraquinone of Rhizoma Polygoni Cuspidati can improve some symptoms like polydipsia,polyphagia and body weight loss of diabetic nephropathy model.②The treatment groups all can significantly decrease the kidney index (P<0.01).③High dosage and middle dosage of anthraquinone can lower serum glucose level after 4 weeks of medication.④Different dosage of anthraquinone can significantly decrease blood urea nitrogen level(p﹤0.05)and serum creatinine level(p﹤0.01).7 Influence on metabolism and hemorheology of syndrome of blood stasis of diabetic nephropathy model①All the dosages of anthraquinone can decrease blood viscosity and plasma viscosity (P﹤0.05).②High dosage and middle dosage of anthraquinone can significantly decrease TG level(p﹤0.05) and high dosage of anthraquinone can significantly decrease LDL level(p﹤0.05).Conclusions1 The possible kidney protection mechanisms of anthraquinone of Rhizoma Polygoni Cuspidati are as follows: lowering serum glucose level, regulating metabolic disorder of blood lipid, decreasing urea nitrogen, inhibiting renal hypertrophia, decreasing microamount of protein excretion, lowering the expression of AGEs, RAGE and Smad3 and lastly, improving hemorheological disorder.2 Anthraquinone of Rhizoma Polygoni Cuspidati is one of the essential components of the early intervention of Rhizoma Polygoni Cuspidati on diabetic nephropathy.
