Wu Mei Wan Immune Regulation Mechanisms In Rats With Ulcerative Colitis

Objective:Ulcerative colitis(UC)is an idiopathic chronic inflammatory bowel disease(IBD).Its main manifestations are diarrhea,bloody mucopurulent stool,abdominal pain and tenesmus.Because the disease is difficult to cure,we have been lacking of effective remedies for treating it.In the past years,the scholars of home and abroad have been intent on the study of finding effective drug.We have a long history and rich experience to treat UC by using Chinese medicine.Wumei pill which has significant effect and little side effect is a common prescription to cure UC.This subject studied treatment of effect and mechanism of Wumei pill by making rat model of UC.In particular,we use Wumei pill to treat UC rats and then we observe the content is high or low about cytokines IL-8 and IL-13 which come from colon tissue of the UC rats.We also observe other indexes such as:Nuclear Factor-κB,Adhesion Molecule,Signal Transducer and Activator of Transcription 6.Methods:In the experiment,eighty four SD rats were randomly divided into six groups,Every group had fourteen rats(seven rats were male,seven rats were female), namely the normal control group,the pathologic control group,the solfasalazine(SASP)group,the high dose group of Wumei pill,medium dose group of Wumei pill,low dose group of Wumei pill.Except the normal control group,the other groups were made through the integration of general immunity and local stimulation (DNCB and acetic acid).These five groups were treated by distilled water, solfasalzine,high dose,medium dose and low dose of Wumei pill respectively.After treatment,the changes of symptoms and immune indexes in the colon tissue such as IL-8,IL-13 gene expression in the colon tissue,the protein expression of NF-κB,CD54,CD44 were observed and detected.The colon pathological changes of every group were also observed under light microscope and transmission electron microscope.Results:(1)The study showed that the weight of rats in model group was lower than that in normal group(P<0.01).The level of IL-8 in pathologic group was significantly higher than that in normal group(P<0.01),and the level of IL-13 was significantly lower than that in normal group(P<0.01).(2)The rats in the model group can been observed the changes of the symptoms such as intermittent diarrhea with mucus and blood at stool,.emaciation,laziness,no luster in hair,poor appetite,loss of weight and increase of colonic damage scores.The colon tissue damaged score in pathologic group is higher than that in normal group (P<0.01).After treatment,their symptoms and sighs obviously improved.The colon tissue damaged score in pathologic group was higher than that in the high,medium,low dose group of Wumei pill and SASP group.There was more difference in local loss of colonic mucous membrane,gland arrangement being regular or not,mucosa hyperemia and inflammatory cell infiltration and so on in each group.(3)The level of IL-8 in pathologic group was significantly higher than that in normal group(P<0.01),and the level of IL-13 was significantly lower than that of the normal group(P<0.01).The level of IL-8 in Wumei pill high,medium,low dose group,SASP group was lower than in pathologic group(P<0.01 or P<0.05),and the level of IL-13 was higher than pathologic group(P<0.01 or P<0.05).The level of IL-8 in Wumei pill high dose group was lower than that in SASP group(P<0.05),and the level of IL-13 was higher than that in SASP group.The level of IL-8 was in Wumei pill low dose group was higher than that in SASP group(P<0.05),and the level lower than that in SASP group.The level of IL-8 and IL-13 in Wumei pill medium dose group was nearly to that in SASP group(P>0.05).(4)The expression of CD44 and CD54 in pathologic group was significantly higher than that in normal group(P<0.01).The expression of CD44 and CD54 in high,medium,low group of Wumei pill and SASP group was lower than that in pathologic group(P<0.01).The expression level of CD44 and CD54 in each Wumei pill group was nearly to that in SASP group(P>0.05).The expression of CD44 and CD54 in SASP group and each Wumei pill group were higherthan that in normal group(P<0.05 or P<0.01).(5)The expression of NF-κB and IL-8 in pathologic group was significantly higher than that in normal group(P<0.01).The expression of NF-κB and IL-8 in high,medium,low group of Wumei pill,SASP group was lower than that in pathologic group(P<0.01).The protein expression level of NF-κB and IL-8 in each Wumei pill group were nearly to SASP group(P>0.05).(6)The expression of STAT6mRNA in pathologic group was significantly lower than that in normal group,The expression of STAT6mRNA in Wumei pill high,medium,low group,SASP group was higher than that in pathologic group.Conclusion:This model was successful.The treatment of Wumei pill was effective on the UC rats.Wumei pill treating the rats with UC has dose-effect relationship.Cytokines IL-8 and IL-13 were involved in the formation of the UC. By increasing the content of IL-13 and reducing the content of IL-8,Wumei pill could recover the balance of cytokines and achieved the purpose of regulating the immune response.Adhesion molecule CD44 and CD54 were involved in the formation of the UC.Wumei pill could decrease the adhesive ability of inflammatory cell,it could regulate immune response.Nuclear factor-κB played an important control role in the formation of the UC.Wumei pill could decrease the expression of NF-κB and IL-8, then it cloud reduce inflammatory damage.Wumei pill could increase the expression level of STAT 6,which regained immune function.
