Gastric carcinoma, as one of the most common malignant tumor in digestive tract, is a polygenetic, multistage, and multicause lesion. The mechanism is still not completely understood. In this study, we investigate the role of wnt signaling passway and related factors such as PP2A, NF-κB in gastric carcinoma progression.Results:1. the results of test 11.1 The results of immunochemistry1.1.1 Immunochemistry staining results of Wnt. The expression of Wnt in carcinoma specimen are stronger than that of control group significantly (P<0.05). 78.8% gastric carcinoma specimen are positive compared to 20% control group. Further analysis show that wnt expression in progressing carcinoma were stronger than that of early stage carcinoma (P<0.05). Wnt expression is correlated to poor differentiation and progression.1.2 Immunochemistry staining results ofβ-catenin The expression ofβ-catenin in carcinoma specimen were stronger than that of control group significantly (P<0.05). Further analysis showβ-catenin expression is correlated to Gastric carcinoma staging. However,β-catenin expression is not correlated to lymph node status.1.1.3 Immunochemistry staining results of APC The expression of APC in carcinoma specimen were weaker than that of control group significantly (P<0.05). Further analysis show APC expression is positively correlated to differentiation (P<0.05).1.1.4 Immunochemistry staining results of c-myc The expression of c-myc in carcinoma specimen were stronger than that of control group significantly (P<0.05).C-myc expression is correlated to Gastric carcinoma progression, poor differentiation and lymph node metastasis.1.1.5 Immunochemistry staining results of NF-κB The expression of NF-κB in carcinoma specimen were stronger than that of control group significantly (P<0.05). Besides, NF-κB expression is correlated to Gastric carcinoma progression, poor differentiation and lymph node metastasis(P<0.05).1.1.6 Immunochemistry staining results of PP2A The expression of PP2A in carcinoma specimen were weaker than that of control group significantly (P<0.05). Further analysis show PP2A expression is positively correlated to Gastric carcinoma differentiation (P<0.05).1.2 Results of RT-PCR1.2.1 RT-PCR results ofβ-catenin In the control group, expression ofβ-catenin mRNA is lower. While in the cancer group, poor differentiated tumors have higherβ-catenin mRNA expression (P<0.05).1.2.2 RT-PCR results of APC In the control group, expression of APC mRNA is higher. while in the cancer group, poor differentiated tumors have higher APC mRNA expression (P<0.05).1.2.3 RT-PCR results of PP2A In the control group, expression of PP2A mRNA is higher .while in the cancer group, poor differentiated tumors have higher PP2A mRNA expression (P<0.05).1.3 Results of western blot1.3.1 western blot results ofβ-catenin In the control group, expression of β-catenin protein is lower. while in the cancer group , poor differentiated tumors have higherβ-catenin protein (P<0.05).1.3.2 western blot results of Wnt In the control group, expression of Wnt protein is lower, while in the cancer group, poor differentiated tumors have higher Wnt protein expression (P<0.05).1.3.3 western blot results of APC In the control group, expression of APC protein is lower, while in the cancer group, poor differentiated tumors have higher APC protein expression (P<0.05).1.3.4 western blot results of PP2A expression of PP2A protein is lower, while in the cancer group, poor differentiated tumors have higher PP2A protein expression (P<0.05).2. The results of test 22.1 Results of RT-PCR2.1.1 RT-PCR results ofβ-cateninβ-catenin mRNA expression of routine cultured SGC-7901 cell line is high. while in the group which interfered byβ-catenin micro RNA,β-catenin mRNA expression is lower (P<0.05).2.1.2 RT-PCR results of APC APC mRNA expression of routine cultured SGC-7901 cell line is low. while in the group which interfered byβ-catenin micro RNA, APC mRNA expression is high (P<0.05).2.1.3 RT-PCR results of PP2A PP2A mRNA expression of routine cultured SGC-7901 cell line is low, while in the group which interfered byβ-catenin micro RNA, PP2A mRNA expression is high (P<0.05).2.2 Results of western blot2.2.1 western blot results ofβ-cateninβ-catenin protein expression of routine cultured SGC-7901 cell line is higher, while in the group which interfered byβ-catenin micro RNA,β-catenin protein expression is low (P<0.05).2.2.2 western blot results of APC APC protein expression of routine cultured SGC-7901 cell line is low , while in the group which were interfered byβ-catenin micro RNA, APC protein expression is high (P<0.05).2.2.3 western blot results of PP2A PP2A protein expression of routine cultured SGC-7901 cell line is low, while in the group which interfered byβ-catenin micro RNA, PP2A protein expression is high (P<0.05).Conclusion1. Wnt signaling pathway plays an important role in the development of gastric carcinoma.2. Wnt signal pathway contains several important proteins, which includesβ-catenin and APC. They play very important roles in progression and differentiation of gastric carcinoma.3. PP2A may regulate the development of gastric carcinoma by the Wnt pathway.4. As an important nuclear transcription factor, NF-κB regulates the progression of gastric carcinoma.
