Objectiveâ‘ To investigate the changes of the expression of IGF-1mRNA,GLP-2,GLP-2RmRNA in intestinal mucosas of experimental obstructive jaundice rats.â‘¡To explore the effects of entemal immunonutrition in the expression of IGF-1mRNA,GLP-2, GLP-2RmRNA in intestinal mucosas of experimental obstructive jaundice rats and the relationship with intestinal mucosa barrier.Methods The experimental obstructive jaundice rats were performed by ligaturing and transecting the common bile duct.The rats randomly divided into five groups:empty group,sham-operated group, common bile duct ligation(CBDL)control group,CBDL+standard enteral nutrition(Nutrison)group,CBDL+enteral immunonutrition(Supportan) group.The rats were killed on the day 5,10,15 to acquire intestinal mucosas and portal vein blood.The expression of IGF-1mRNA and GLP-2RmRNA of rats' intestinal mucosas was detected by using RT-PCR method.The expression of GLP-2 of rats' intestinal mucosas was detected by using Westem-Blotting method.The apoptosis of rats' intestinal mucosas was detected by using TUNEL methods.The endotoxin concentration in rats' portal vein blood was detected by limulus test.The data of all groups were collected and analysed.Results The expression of IGF-lmRNA and GLP-2RmRNA in intestinal mucosa was down-regulated in CBDL control group,but the expression of GLP-2 did not change.The intestinal mucosa apoptotic rate and the portal vein blood endotoxin concentration were higher in CBDL control group than in empty group or sham-operated group.The expression of IGF-1mRNA,GLP-2 and GLP-2RmRNA in CBDL group rats' intestinal mucosa were up-regulated by enteral immunonutfition (Supportan)and standard nutrition(Nutrison).The intestinal mucosa apoptotic rate and portal vein blood endotoxin concentration were reduced by Supportan and Nutrison.Conclusionsâ‘ The down-regulation of IGF-lmRNA and GLP-2RmRNA in intestinal mucosa may connect mith the intestinal mucosa barrier injury ofexpenimental obstructive jaundice rats.â‘¡The up-regulation of IGF-lmRNA,GLP-2 and GLP-2RmRNA in obstructive jaundice rats' intestinal mucosa by standard nutrition(Nutrison)and enteral immunonutrition(Supportan)may be one of the mechanism to improve the intestinal mucosa barrier.â‘¢The enteral immunonutrition has stronger effects than standard enteral nutrition in the expression of IGF-lmRNA,GLP-2 and GLP-2RmRNA in the intestinal mucosas of obstructive jaundice rats.
