Experiment Research Of Shenshuaixiezhuo Pill On Effecting BMP-7/Smads/TGF-β1 Signal Transduction Pathway In Chronic Renal Failure Rats

Objective:To investigate the effect of the Shenshuaixiezhuo Pill to kidney function and kidney pathologic tissue of adenine caused chronic renal failure rats and BMP-7/Smads/TGF-β1 signal transduction pathway,and to illustrate the mechanism of Shenshuaixiezhuo Pill on delaying chronic renal failure and relieving renal interstitial fibroblast.Methods.In the experiments,the adenine caused chorinic renal failure model is adopted.It is observed for the change of the common physical order,weight,BUN,Scr and Hb in blood serum ofexperimental rats.HE and Masson staining method for pathologic slice are used to observe the pathlologic change and kidney fibrosis of the experimental rats kidney tissue afer therapied.The protein expression of TGF-β1,Smad2/3,Smad6,BMP-7, BMP-7RⅡin kidney tissue are detected respectively by immunohistochemistry. The proteins expression of Smad2/3,Smad6,BMP-7,BMP-7RⅡare also detected by Western Blot method.The expression of TGF-β1mRNA, BMP-7mRNA and BMP-7 receptor-ⅡmRNA are also detected by semi-quantitative RT-PCR.Results:The common physical order and weight of the CRF rats are improved obviously after being treated by Shenshuaixiezhuo Pill,BUN and Scr in blood serum is reduced which has the significant difference compared with antithesis groups and Hb in blood serum is improved which hasn't the significant difference compared with antithesis groups.The Shenshuaixiezhuo Pill can adjusts the TGF-β1 and Smad2/3 protein to low expression and the Smad6,BMP-7 and BMP-7RⅡto high expression in kidney tissue,which has the significant difference compared with antithesis groups.The expression of TGF-β1mRNA are also reduced and the expression of BMP-7mRNA,BMP-7RⅡmRNA are creased,which havesignificant difference with antithesis groups.Conclusions:The Shenshuaixiezhuo Pill can possibly block TGF-β1/ Smads signal transduction pathway by reducing TGF-β1 and Smad2/3 protein expressions and creasing Smad6,BMP-7 and BMP-7RⅡproteins expression in kidney tissue.It is maybe one of mechanisms of Shenshuaixiezhuo Pill on delaying chronic renal failure that Shenshuaixiezhuo Pill can relieve renal interstitial fibroblast by influning BMP-7/Smads/TGF-β1 signal transduction pathway.
