ObjectiveTo study the protective effects and mechanisms of joint use of selenium and sodium tanshinone IIA on myocardial ischemia reperfusion in rabbits.MethodsTo set up rabbit model of myocardial ischemia reperfusion (MIR),the levorotary cornary artery was ligated for 40 minutes and reperfused for 60 minutes.Twenty-seven rabbits were randomly divided into four groups,namely non-operation group,ischemia reperfusion group (IR),sodium tanshinone IIA(DS-201) group and joint use of selenium and DS-201group.At the end of 60 minutes reperfusion,the blood was gathered and the heart was taken off. The lactate dehydrogenase (LDH) and creatine kinase (CK) activity in plasma were detected.The superoxide dismutase (SOD),glutathione peroxidase (GSH-Px)activity and malondialdehyde (MDA) content in plasma, myocardial tissue and myocardial mitochondria were detected.The content of nitric oxide (NO)in plasma,myocardial tissue and the activity of total nitric oxide synthase(T-NOS),inducible nitric oxide synthase(iNOS) in myocardial tissue were detected.After total lipids in myocardial tissue being extracted,the phospholipids content of membrane lipids and total cholesterel content in myocardial tissue were detected. The cardiolipin content in myocardial tissue was detected through the method of thin-layer chromatography analysis.Results①In joint use of selenium and DS-201group,the LDH and CK activity in plasma were significantly lower than that in group DS-201(P<0.01)alone. In DS-201group LDH and CK activity in plasma were significantly lower than group IR(P<0.01).②In group IRI, the period of two next P waves was significantly prolonged(compared with the period of ligation before, P<0.01), S-T segment was merged into T wave and became a unidirectional curve after the ligation of levorotary cornary artery. The period of two next P waves was further significantly extended and S-T segment significantly rose further during the reperfusion of levorotary cornary artery. In joint use of Selenium and DS-201group, the changes of the period of two next P waves and S-T segment were the same as group IRI. During the reperfusion the period of two next P waves was no significantly prolonged compared with the period of the ligation. S-T segment was significantly declined during the reperfusion period(compared with group IRI, P<0.01).③In joint use of selenium and DS-201group,the activity of GSH-Px in plasma, myocardial tissue and myocardial mitochondria were significantly higher than that in group DS-201 alone(P<0.05 or P<0.01),group IR(P<0.01)and non-operation group(P<0.05). But there were no significant differences of activity of GSH-Px in plasma, myocardial tissue and myocardial mitochondria between group DS-201 and group IR.④In joint use of Selenium and DS-201group and DS-201group,the activity of SOD in plasma, myocardial tissue and myocardial mitochondria were significantly higher than that in group IR(P<0.05 or P<0.01).But there were no significant differences of activity of SOD in plasma, myocardial tissue and myocardial mitochondria between joint use of selenium and DS-201 group and DS-201group.⑤In joint use of selenium and DS-201group,the MDA content in plasma, myocardial tissue and myocardial mitochondria were significantly lower than that in DS-201group(P<0.05 or P<0.01)alone.In group DS-201the MDA content in plasma, myocardial tissue and myocardial mitochondria were significantly lower than group IR(P<0.05 or P<0.01).⑥In joint use of selenium and DS-201group,the phospholipids content of membrane lipids and cardiolipin content in myocardial tissue were significantly higher than that in DS-201group alone(P<0.05). The total cholesterel content in myocardial tissue was significantly lower than that in DS-201group alone(P<0.01). In DS-201group the phospholipids content of membrane lipids and cardiolipin content in myocardial tissue were significantly higher than that in group IR(P<0.05).⑦In joint use of selenium and DS-201group and DS-201group,the content of NO in plasma, myocardial tissue were significantly higher than that in group IR(P<0.05 or P<0.01).But there were no significant differences between joint use of selenium and DS-201group and DS-201group.⑧In joint use of selenium and DS-201 group and DS-201group,the activity of T-NOS in myocardial tissue was significantly higher than that in group IR(P<0.05 ).But there was no significant difference between joint use of selenium and DS-201group and DS-201group.⑨The activity of iNOS in myocardial tissue was no significant difference in four groups.Conclusions①In joint use of selenium and DS-201group,the LDH and CK activity in plasma were significantly lower than that in group DS-201(P<0.01)alone.②In joint use of Selenium and DS-201group, during the reperfusion the period of two next P waves was no significantly prolonged compared with the period of the ligation. S-T segment was significantly declined during the reperfusion period(compared with group IRI, P<0.01).③In joint use of selenium and DS-201group,the activity of GSH-Px in plasma, myocardial tissue and myocardial mitochondria were significantly higher than that in group DS-201 alone(P<0.05 or P<0.01).④In joint use of Selenium and DS-201group,the activity of SOD in plasma, myocardial tissue and myocardial mitochondria were significantly higher than that in group IR(P<0.05 or P<0.01).But there were no significant differences of activity of SOD in plasma, myocardial tissue and myocardial mitochondria between joint use of selenium and DS-201 group and DS-201group.⑤In joint use of selenium and DS-201group,the MDA content in plasma, myocardial tissue and myocardial mitochondria were significantly lower than that in DS-201group(P<0.05 or P<0.01)alone.⑥In joint use of selenium and DS-201group,the phospholipids content of membrane lipids in myocardial tissue were significantly higher than that in DS-201group alone(P<0.05).
