The geoherbalism of Chinese crude drug is the traditional objective method to evaluate herbs,and its quality character and forming mechanism are the main content of present research.It is the key to study geoherbalism connotation by modern and scientific methods.Licorice was studied as an example in this thesis. The development and evolutionary process of licorice geoherbalism were studied. In order to find the material connotation,different licorices with variable characters,habitats,species were researched with fingerprint and multiple constituents assaying.A reasonable evaluation system of genuine licorice was discussed.The results were as follows:1.The habitats of licorice in history were different,but the characters were relatively stable.Gansu,Ningxia,Neimeng,Shannxi and Shanxi were recorded as the genuine habitats,and brownish red,strong,transverse striation,complexion yellowish and abundant flouring were considered as the good characters with high quality.The traditional species was Glycyrrhiza uralensis Fisch.2.The reference substances of glycyrrhizin,liquiritin,licochalcone A and isoliquiritigenin with above 98%purity were separated and prepared through the method of macroporous resin,high speed counter currents chromatographic technique,reversion phase chromatography and gel chromatography.The HPLC-DAD and RRLC-MS/MS method were estabished for the study of fingerprint and simultaneous determination of glycyrrhizin,liquiritin,licochalcone A,isoliquiritigenin,glycyrrhetic acid and fermononetin.3.The contents of multi-index compositions in different character samples were variable.The contents of glycyrrhizin,liquiritin and isoliquiritigenin in brownish red,complexion yellowish and abundant flouring samples were higher than the matching characters.As a result,these components might be considered as the main geoherbalism index of licorice.4.The fingerprint and contents of multi-index compositions in samples from different habitats were variable.Licorice from Xinjiang was different from other habitats significantly and the similarity of fingerprint is 0.400-0.700.The habitat conditions were important for the geoherbalism.5.The fingerprint of three licorice species was different.The contents of multi-index compositions in samples were variable.Besides other constituents, liquiritin and licochalcone A could be considered as the index components to discriminate the different species.The species conditions were also important for the geoherbalism.6.A reasonable evaluation system of genuine licorice was raised.The main character index were yellowish and abundant flouring complexion.Traditional producton places were the good habitat index of genuine licorice.Glycyrrhizin, liquiritin and isoliquiritigenin were the main chemical index.The research characteristic and innovation of this thesis are as follows:(1) The multiple view angers of literature research,traditional character and chemical charater of licorice geoherbalism were studied.(2) The method of simultaneous determination of glycyrrhizin,liquiritin,licochalcone A,isoliquiritigenin, glycyrrhetic acid and fermononetin were found with HPLC-DAD and RRLC-MS/MS.(3) The main substances fundament of genuine licorice were identified with the method of comparing different characters,habitats and species licorice samples,and a reasonable evaluation system of genuine licorice was raised...
