This dissertation investigated chemical constituents of ethanolic extracts of the root of Adina polycephala and the stem bark of Fraxinus sieboldiana and their bioactivities on several pharmacological models. By using a variety of chromatographic techniques, 50 compounds were isolated, 30 (A1*-A30) from A. polycephala and 20 (F1*-F20) F. sieboldiana. On the basis of spectroscopic and chemical methods, their structures were determined. Their chemical names and structures are listed in Table 1 and Figure 1.Among them, 17 structures are new, including four dimeric phenolic glycosides (A1*-A4*) and a new iridoid diglycoside (5*) from A. polycephala, as well as seven abietane diterpenes (F1*-F7*) and five nor-abietane diterpenes (F8*-F12*) from F. sieboldiana. At a concentration of 10-5M, A3*, F7*, F10* and F13 showed potent in vitro activities against the release of glucuronidase in rat polymorphonuclear leukocytes induced by platelet-activating factor (PAF), with inhibitory rates of 60.2%, 59.7%, 56.1% and 65.9%, respectively. A13 exhibited nuroprotective activity against serum deprivation-induced PC 12 cell apoptosis with protection ratio of 51%. Compound F14 was active against H5N1 avian influenza virus with IC50 value of 4.8μM. In addition, A4*, F1* and F6* showed selective cytotoxicity against several human cancer cell lines including lung (A549), ovarian (A2780) and lung (A549) cancer cells, with IC50 value of 0.21, 1.72 and 6.04μM.Table 1.
