Background and ObjectiveIntimal hyperplasia and thrombotic occlusion of small caliber artificial vessels after transplanted,causing failures,has been attributed to lacking of the endothelial layer on the surface of the artificial vessels.It has confirmed that quick endothelialization of the artificial vessels can prevent or at least decrease the rates of restenosis.People have investigated the endothelialization from seeding endothelial cells on the surface of artificial vessels to delivering genes.Vascular endothelial growth factor(VEGF) is the most specific factor promoting endothelial cell proliferation.However,both coating with VEGF protein and transfecting genes with plasmid have their limitations.Adenovirus,as a gene carrier,has showed the effective transfecting talent.Accordingly,the present investigation was designed to test the hypothesis that artificial vessels carting adenovirus-mediated vascular endothelial growth factor could transfect endothelial cells,produce VEGF proteins,and promote endothelial cells proliferation.Methods and ResultsBy using ad5-EGFP,the optimal MOI for adenovirus vector to infect endothelial EAhy926 cells was determined at 200.The artificial vessels carring adenovirus released virus vector gradually in the static water. Scanning electronic microscopy showed that Endothelial cells could adhere on the surface of the artificial vessels.RT-PCR had detected the VEGF gene of the ad5-VEGF165 transfected groups,which was more significant on day 3 than day 1,the control groups hadn't detected.Using ELISA kits to measure the VEGF proteins of the cell culture supernate, the ad5-VEGF165 transfected groups growed grandually and reached the peak by day 5,then began to drop down.The concentrations of control groups were very low.The endothelial cells proliferated greatly of the ad5-VEGF165 transfected groups,which was slowly of the control groups(P<0.01).ConclusionsArtificial vessels earring adenovirus-mediated vascular endothelial growth factor can transfect endothelial cells,produce VEGF proteins,and accelerate endothelial cells proliferation.
