ObjectiveTo investigat the expression of galectin-9 and Tim-3 in mice nasal mucosa and to reveal the relationship with the pathogenesis of allergic rhinitis.MethodsFourty mice were divided into 4 groups:AR group,Rosiglitazone group,Dexamethasone group,and normal group,10 mice in each group.On days 1 and 14,mice were immunized by ovalbumin(OVA).From d 21to 27,the sensitized mice were exposed to OVA each day.In this study,the mice of Group Rosiglitazone and Group Dexamethason received the test drugs from day 21 to day 27 after the challenge.Mice in normal group were exposed to the saline.Serum was collected on day 27.We observatde the symptom of the mice such as sneezing,nasalrubbing and nasal discharge.And then,we killed the animal.Immunohistochemical staining was uesed to examine the galectin-9 and Tim-3 expression in nasal mucosa.RT-PCR was performed to investigate the level of mRNA expression of complement galectin-9 and Tim-3 in nasal mucosa.ELISA was performed to investigate the level of proteinum expression of IL-4 (interleukin-4),IL-5 and IFN-γ(interferon-γ) in serum.ResultsGalectin-9 and Tim-3 were detected in both groups.Expression of galectin-9 and Tim-3 in group AR was higher than that of group N.Dex and ROS can down regalate the expression of them.Pearson correlation analysis demonstrated that the level of galectin-9 and Tim-3 was positivly correlaed with the level of IFN-γ,but negativly correlated with the level of IL-4 and IL-5.Conclusion Galectin-9 and Tim-3 may play an important role in the pathogenesis of allergic rhinitis.
