Object:To observe the Influence of Zhi-Bai-Di-Huang Decotion on Ureaplasma urealyticum(UU)-infected rats'spermatogenic cell apoptosis and effect factors Caspase-3 and Caspase-9 of mitochondria expressions.Method:45 out of 60 male SD rats were randomly drawed and made into the UU infected animal models,the rest of which were the sham operation group. The UU infected animal models were then divided randomly into the model group, the minocyclin group and Zhi-Bai-Di-Huang Decotion group,each with 15 rats and dosaged with lavage.The model group and the sham operation group were lavaged with isotonic Na chloride,,while the minocycline group and Zhi-Bai-Di-Huang Decotion group were lavaged with the corresponding medicine. After 21 days, the rats were killed and the testicle weight,the UU cultivation results, the apoptosis rate of reproductive cells, Caspase-3 and Caspase-9 expressive level,the pathologic manifestations of the testicles,and ultramicrostructure change of spermatogenic cells of each groups were measured and compared.Result:There was statistical meanings in the differences of the testicle weight,the positive rate of the UU cultivation results, the apoptosis rate of reproductive cells, Caspase-3 and Caspase-9 expressive levels,the classifications of the pathologic manifestations of the testicles (P< 0.05) between the model group, the sham operation group,the minocycline group and the Zhi-Bai-Di-Huang Decotion group, so was the last three groups, while not between the minocycline group and the Zhi-Bai-Di-Huang Decotion group.Conclusion:1. Zhi-Bai-Di-Huang Decotion can actively depress the growth and production of UU and is anti-UU.2.UU infection can lead to the increasing of spermatogenic cell's apoptosis in rats.It is proved that the over-apoptosis of spermatogenic cell is the important mechanism of the descending quality of the sperms after UU infection which leads to sterilization.3.The stronger expressive level of Caspase-3. Caspase-9 in spermatogenic cells in UU infected rats explained that they are the important factors which promots the cell apoptosis,and mitochondria plays a key role in the regulation to spermatogenic cells.4. Zhi-Bai-Di-Huang Decotion can down regulate the expressive level of Caspase-3 and Caspase-9.In other words,one of the mechanisms of Zhi-Bai-Di-Huang Decotion treating UU infection and improving the sperm quality is to anti-apoptosis through regulate the expressions of the apoptosis effect factors Caspase-3 and Caspase-9...
