Objective:To study protective mechanisms of superficial needling to ischemic penumbra neuron in cerebral infarction rats.Methods:72 SD rats were randomly divided into superficial needling group(Groupâ… ),routine needling group(Groupâ…¡),model control group (Groupâ…¢),normal control group(Groupâ…¢).The first three groups were established as the cerebral infarction models with thromboembolism. Groupâ…£was tied up and fixed at the same time without any processing factors.Each group was divided again at postoperative 2days,7days 14 days three time points,each time points 6 rats.Observe the proliferation, regeneration and apoptosis with the rat brain neuron after 2,7,14 days in the treated Groupâ… ,â…¡and the untreated groupâ…¢,â…£in the corresponding time from the neurological score,brain pathological changes determined by HE staining,Caspaase-3,bFGF,NGF protein expression changes in brain tissue measured by immunohistochemistry method.It has remarkable social and practical significance to reveal the protective mechanisms of superficial needling to ischemic penumbra neuron in cerebral infarction rats in the view of cerebral nerve function reconstruction,modern immunology and molecular biology. Result:1.Superficial needling can effectively improve neurological defect sign in cerebral infarction rats.2.Caspase-3 protein expression in brain tissue ischemic penumble rats of the superficial needling group and routine needling group were significantly lower than that of the model control group in the three time points.3.NGF,bFGF expression in brain tissue ischemic penumble rats of superficial needling group and routine needling group were significantly higher than that of the model control group in the three time points.4.Caspase-3 protein expression in brain tissue ischemic penumble rats of the superficial needling group was lower than that of routine needling group,while NGF,bFGF expression was higher than that of the routine needling group in the three time points.5.Increasing blood NGF,bFGF expression level in ischemic penumbra neuron in cerebral infarction rats after superficial needling,can interfere the mechanism of apoptosis by preventing the overload of calcium ions, reducing free radical lesion and resisting excitatory amino acid toxicity, then reduce caspase-3 protein expression,prevent the activation of caspase-3,prohibit the apoptosis of neuron,and promote the proliferation and regeneration in injured zone. Conclusion:The study confirmed that protective mechanisms of superficial needling to ischemic penumbra neuron might be related to increasing blood NGF,bFGF expression level after neurological defect, extending its high expression time-course,inhibiting the apoptosis of Caspase-3 activated neuron.Superficial needling group was superior to normal group in increasing NGF,bFGF expression,inhibiting Caspase-3 expression.It reveals that meridian has its material basis on the body surface and superficial needling has stimulating effect to make the shallow tissue show its meridian specificity.
