| Radix aconiti lateralis preparata(RALP) is one of commonly effective toxic Chinese herbs, and it's important effect is replenishing yang and saving the inverse.It's toxic components are double-ester alkaloids represented by aconitine. Double-ester alkaloids will hydrolysis into monoester alkaloids and the acinine,and it's detoxification mechanism has been initially set out.From 20th century 90's,study on RALP's compatibility and safety is forefront and hot.Study on relationship between changing compatibility environment and toxicity is key scientific problems about the RALP's applied basic research.Based on the UV,TLC,HPLC,GC,fingerprint,HPLC/MS,macroporous resin,reversed-phase column chromatography and other modern separation purification technology, black prepared lateral root of aconite(BPLRA) and Radix Glycyrrhizae are selected to study its toxic components of the dynamic process in the compatibility environment, at the same time, combined-line-detection,intelligent'multi-column,tracking of active indication,components-oriented,technology has been innovatively applied.To track its hydrolysis product and material basis related to anti-arrhythmia,and to study on regulating RALP's toxicity mechanism of changing compatibility environment,Conclusions were obtained as follows:In the internal-compatibility-environment:Mg2+,MA, BP-5 and other components are material basis related to anti-toxic in the internal-compatibility-environment,which may exert bi-directional regulating the biological effects through the amount of its material.In the external compatibility environment:monoester alkaloids are material basis of toxic constituents, and its compatibility is mainly affected by the characteristic dose,pH,time,metal ions and so on. liquiritin and Ammonium Glycyrrhizinate exert cooperative inhibitory effect through the synthesis of salt changing the form of alkaloids. Glucose,salt,starch has a role to accelerate the hydrolysis.Acidic environment is the key factors which restrain aconitine hydrolysis into monoester alkaloids, namely can decide toxic ingredients hydrolysis manner by changing the composition of the positive effect of nitrogen atoms and space structure.Regulation hypothesis of changing compatibility environment reveals that changing compatibility environment will regulate toxicity of the material basis, the key factors and mechanism, approaching the sciencity and rationality of compatibility disciplines.SO basicly sets out the scientific connotation of regulating RALP's toxicity mechanism of changing compatibility environment, at the same time, provides a new ideas of studying on formula of TCM. |
