Radix Aconiti Lateralis Praeparata is the most effective medicine and widely used in clinic on reinforcing fire and yang. Today, just four kinds of prepared slices, heishunpian, baifupian, danfupain and paofupian, have been recorded in Pharmacopoeia of the People’s Republic of China. But there is no more study on danfupain and the influence on nifuzi processing in fuzi-producing region. So we observed the appearance and text the contents of diester alkaloids, monoester alkaloids and benzoic acid in danfupain and prepared slics of shengfupian to study on significance of processing with auxiliary materials. And we also built feature print for assisted analysis.The research includes four parts, and the results are as follows:Part 1 OverviewThe auxiliary materials processing methods of Radix Aconiti Lateralis Praeparata had been collected. Especially the history, usage, TCM theories of Black beans of Radix et Rhizoma Glycyrrhizae processing method. The key points in preparing were also found. The compositions, including the diester alkaloid, the monoester alkaloid, the lipidic alkaloids, the amino alcohol alkaloid, were collected and classified. Pharmacological researches showed that Radix Aconiti Lateralis Praeparata have heart, nerve and precreation toxicity. On the other hand, it has a lot of pharmacological effects, too, for example, improving cardiovascular system founction, anti-inflammation, analgesia and improving energy metabolism. In addition, for further information, the influence of Radix et Rhizoma Glycyrrhizae on Radix Aconiti Lateralis Praeparata ether in constituents or in pharmacological effects, when they are used together, were summarized.Part 2 Experimental Study1. Measures and study on detection methodTraditional trait and ingredients detection were chosen as measures to study the influence of Radix et Rhizoma Glycyrrhizae and black beans on Radix Aconiti Lateralis Praeparata. Trait included color, character, order and taste. Ingredients detection included the content of diester alkaloids, the content of monoester alkaloids and HPLC feature print. Detection of diester alkaloids and monoester alkaloids was built on the basis of the methord on Pharmacopoeia. And HPLC feature print was built for more constituents study.2. Study on danfupian preparing technicsThe different degree prepared slices were prepared to study the two key points in danfupian processing. The first one is blanching until no salt. The second one is boiling until no white core and without any water.The first step of the processing is blanching yanfuzi until all salt is rinsed out. The time of blanching process is relative to temperature. Higher the temperature is, quicker the salt rinsed out. The contents of three diester alkaloids, three monoester alkaloids and benzoic acid and recovery rate were reducing in order of inferior, moderate, overage praparion. The contents of three diester alkaloids, three monoester alkaloids and benzoic acid in moderate praparion have reduced by 74%,57% and 78%. It is the best for without salt and has more efficient ingredients.The second step of the processing is boiling with auxiliary materials until white core turn to brown and without any water. The of three diester alkaloids, three monoester alkaloids and benzoic acid in moderate praparion were higher than the inferior ones. So it is necessary to boil moderately.Blanch yanfuzi with 30 times water,2 times a day until all salt is rinsed out. That is controlled by salinity meter. And boil with auxiliary materials until white core turn to brown and without any water.3. Shengfupian processing with black beans and Radix et Rhizoma GlycyrrhizaeShengfupian, a Chinese Materia Medica just been dried after being harvested, was used in this study, to study the ancient auxiliary materials processing method. Ancient auxiliary materials processings used fresh Radix Aconiti Lateralis Praeparata, which was different from today. So the technics of processing with shengfupian was study.Soaking could influence appearance and lose ingredients. When boiling the same time, the production of with soaking was not without white core. The content of diester alkaloid in the production boiling with soaking water is 0.0081%-0.012%, which may be higher than the bound. The processed product could boil to no white core in 2 hours, and was not spicy and hot at all in 3 hours. The content of diester alkaloids were reducing, while monoester alkaloids were rising in 3 hours. Both soaking and boil time could impact the quality of processed slics. So Shengfupian should be processed by soaking until without dry core, and boiling 2 hours with clear water.4. The scientific purpose of blanched yanfuzi boiling with black beans and Radix et Rhizoma GlycyrrhizaeProcessing with black beans and Radix et Rhizoma Glycyrrhizae could reduce the contens of three diester alkaloids, three monoester alkaloids and benzoic acid, and bring in new constituents. The result was concluded by conbining apprence, detection results and feature prints. There are 15 peaks in danfupian’s feature print. The ingredients of yanfuzi are mainly before 31 min, while the ingredients of black beans and Radix et Rhizoma Glycyrrhizae are mainly after 31 min under the detection wavelength of 270 nm. When the feature is detected by HPLC-MS,116 signals were found in positive ions mode, of which 37 signals could be found in yanfuzi,19 signals could be found in Radix et Rhizoma Glycyrrhizae,11 signals could be found in black beans. And for Radix et Rhizoma Glycyrrhizae, there were 21 signals found in in negative ions mode. Processing with black beans and Radix et Rhizoma Glycyrrhizae could change the content and material composition the of danfupian. But in order to learn the further scientific purpose of blanched yanfuzi boiling with black beans and Radix et Rhizoma Glycyrrhizae, more studies are needed.5. Comparion of Shengfupian processing with black beans and Radix et Rhizoma Glycyrrhizae with yanfuziThe two kinds prepared slices are differernt in apperence, contents and material composition. The content of every component in Shengfupian processing with black beans and Radix et Rhizoma Glycyrrhizae was higher than danfupian. That may has relation with quantity of water treatment to yanfuzi and danfupian. After processing the content of diester alkaloid was all reducing, and the content of monoester alkaloid and benzoic acid was all rising in both Shengfupian and yanfuzi prepated slics. And the ingredients of black beans and Radix et Rhizoma Glycyrrhizae were taken to both Shengfupian and yanfuzi prepated slics. The influence on constituents of auxiliary materials was same, but further study was needed,In summary, processing with auxiliary materials could make a great difference with boiling with water only. The content of every conponient was lower, and more constituents were detected. But the relationship between the changing and pharmacological effects need a further research.
