Affect And Correlation Of Chronic Atrophic Gastritis And Precancerous Lesions Associated Genes And Mechanisms Treated By Huazhuo Detoxification

Part one:The Expression and Correlation of CyclinD1 and PTEN in Multi-stage of Gastric CancerObjective:To investigate the expression and relevance of CyclinD1 and PTEN in multi-stage of gastric cancer by immunohistochemical staining. Not only to study the mechanism of gastric cancer to provide a theoretical basis for the evidence for the early diagnosis of gastric cancer, but also to find the appropriate combination of Traditional Chinese Medicine and Modern Medicine.Methods:The expression and relevance of CyclinD1 and PTEN on 223 cases gastric cancer specimens were detected at paraffin-embedded specimens of different gastric lesions , which were not treated or not chemotherapy at Chinese Medicine Hospital of Hebei Medical University in 2004 -2008.Results:1. It was statistically significant that the expression of CyclinD1 was increased and the expression of PTEN decreased in chronic superficial gastritis, chronic atrophic gastritis, intestinal metaplasia, dysplasia and gastric cancer gradually .(P <0.05).2. The positive expression of CyclinD1 and PTEN was negatively correlated in gastric cancer multi-stage .Conclusion:1. The positive expression of CyclinD1 is increasing in the progress of gastric cancer of stage, on the other hand, the expression of PTEN decreasing.2. The expression of CyclinD1 protein and PTEN were associated with close level of medium in the progress of gastric cancer of stage, with the increased expression of PTEN protein ,CyclinD1 protein expression decreased.Part two:The study of treatment with patients on precancerous lesions of gastric cancer based on"zhuodu"Objective:Observation the therapeutic effect of the prescription of huazhuo jiedu hewei based on "zhuodu" theory in chronic atrophic gastritis (CAG) precancerous lesions .Methods:110 cases of precancerous lesions of the CAG were randomly divided into treatment group (55 cases) to be given prescription of oral Huazhuo detoxification and stomach prescription, the control group (55 cases) were Weifuchun tablet for 6 months. Observed clinical endoscopy, pathology and treatment efficacy CyclinD1 and PTEN expression before and after the change integral value.Results:Endoscopic mucosal treatment group preclusion and bile reflux improved than the control group, pathological changes of more effective the treatment group than in the control group was high, immunohistochemistry showed Huazhuo detoxification and stomach prescription can only decreased the CyclinD1 expression, increased PTEN expression (P <0.05)Conclusion:1. It was reduced on congestion, edema, erosion, and preclusion of mitigation mucosa, granular hyperplasia and bile reflux in astric mucosa of patients treated by Huazhuo detoxification prescription.2. Huazhuo detoxification and stomach prescription can be able to improve Cure rate on IM, ATP and other different pathological type in patient.3. Huazhuo detoxification and stomach prescription can be able to inhibit the expression of CyclinD1 protein, increased the expression of PTEN protein and inhibit cell proliferation and promoted apoptosis, thus restoring the balance between the cell proliferation and apoptosis.Part three: The study of treatment with Detoxification and stomach prescription on gastric precancerous lesions of gastric cancer related genes in rat based on"zhuodu"Objective:To observe the expression on the gene mRNA of CyclinD1 and PTEN in chronic atrophic gastritis treated by Huazhuo detoxification and stomach prescription in gastric mucosal precancerous lesions of chronic atrophic gastritisMethods:66 Wistar rats were randomly divided into 6 groups 11: normal control group (control group), model group, RA group (RA group), Huazhuo detoxification and stomach prescription with high dose group (high dose group), Huazhuo detoxification and stomach prescription dose treatment group (middle dose group), Huazhuo detoxification and stomach prescription with low dose group (low dose group). Insert spring with Helicobacter replicated with CAG precancerous lesion complex model, the group given medicines administered 1 time per day, each 2mL, for 12 weeks and then the reverse transcriptase gene (RT) and polymerase chain amplification ( PCR) combined (RT-PCR) detection of precancerous lesions and gastric mucosal expression of CyclinD1 and PTENmRNAResults : Compared with the control group, the expression of CyclinD1mRNA in model group was increased, the expression of PTENmRNA was decreased significantly (P <0.05); It was statistically significant difference between the treatment group and model group (P <0.05); high-dose group and RA group was decreased in CyclinD1mRNA, higher than the expression of PTENmRNA in traditional Chinese medicine and the low dose group (P <0.05).Conclusion:1. It had generally improved on the experimental rats treated by Huazhuo detoxification and stomach prescription with chronic atrophic gastritis diet, activity, mental state, hair, feeding, stool and weight;2. Huazhuo detoxification prescription can only improve the pathological changes on experimental gastric chronic atrophic gastritis rats.3. The expression of CyclinD1mRNA with Huazhuo detoxification and stomach prescription was decreased, the expression was significantly higher PTENmRNA.4. The expression on the genes of CyclinD1mRNA and PTENmRNA may be the material to test evidence-based prescription of chronic atrophic gastritis and precancerous lesions.Part four:Detoxification and stomach precancerous lesions in rats on the mechanism of angiogenesisObjective:In this study, to observe Affect on gastric mucosa of VEGF and HIF-1αwith Huazhuo detoxification and stomach prescription in chronic atrophic gastritis and gastric precancerous lesions, and to explore the possible therapeutic mechanism of Huazhuo detoxification prescription in chronic atrophic gastritis and precancerous lesions Angiogenesis area. Methods:66 Wistar rats were randomly divided into 6 groups 11: normal control group (control group), model group, RA group (RA group), Huazhuo detoxification and stomach prescription with high dose group (high dose group), Huazhuo detoxification and stomach prescription dose treatment group (middle dose group), Huazhuo detoxification and stomach prescription with low dose group (low dose group). Insert spring with Helicobacter replicated with CAG precancerous lesion complex model, the group given medicines administered 1 time per day, each 2mL for 12 weeks. then to observe the affect of VEGF and HIF-1αon gastric mucosa with Huazhuo detoxification and stomach prescription in chronic atrophic gastritis gastric mucosa precancerous lesions by immunohistochemistry and western-blot method.Results:Compared with the control group, the expression of VEGF and HIF-1αwas increased in model group and the treatment group, which was increased significantly (P <0.05); the treatment group and model group, the difference was statistically significant (P <0.05); high-dose group and retinoic acid were decreased expression is better than medicine and the low dose group (P <0.05).It was statistically significant difference between the treatment group and model group (P <0.05); high-dose group and RA group was decreased in VEGF and HIF-1αin traditional Chinese medicine and the low dose group (P <0.05), Low-dose group compared with the retinoic acid group showed no significant difference (P> 0.05) Conclusion:1. Huazhuo detoxification and stomach prescription can decrease the expression of HIF-1αand VEGF.2. The expression on the genes of HIF-1αand VEGF may be the material to test evidence-based prescription of chronic atrophic gastritis and precancerous lesions.3. Huazhuo detoxification and stomach prescription may be through anti-angiogenesis mechanism to achieve the prevention and treatment of atrophic gastritis and gastric precancerous lesions, and there is a certain dose of the treatment effect.
