Aims:To research the effects of 10-week high-fat diet on rats as well as a single bout of exercise and long-term exercise on obese rats adipose tissue hormone sensitive lipase(HSL) activity,protein and gene expression;perilipin protein and gene expression;peroxisome proliferator activated receptor-γ(PPAR-γ) gene expression as well as blood fat,weight et al.Methods:116 3-week-age Sprange-Dawley male rats were randomly divided into high-fat group(n=110)and general diet control group(C,n=6).After 10 weeks,66 obese rats(OB group) and 44 obesity resistant rats(OR group) were differentiated.Then the OB rats were randomly divided into: non-exercise control group(EC),immediate post-exercise group(E0) as well as 1(E1),3(E3),12(E12),and 24(E24) hours after exercise group; long-term control group(LC) and long-term exercise group(LE).The concentrations of blood fat,blood sugar,serum free fatty acids(FFA), serum glycerol,adipose tissue HSL activity of obese rats were tested using chemical method.Then the HSL and perilipin protein expressions of rats adipose tissue were tested with Western Blotting method,and the HSL,perilipin and PPAR-γgene expressions of rats adipose tissue were tested with Real-Time PCR method.Results:1.After 10-week high-fat diet,the weight,fat weight,serum FFA and serum glycerol of obesity group(OB group) rats were significantly higher than general diet control group(C group) and/or obesity resistance group (OR group);OB group rats' food availability was higher than OR group or C group(P<0.01).While,the OB group rats adipose tissue HSL activity, HSL protein content as well as gene expression were markedly lower than C group(P<0.05).And the OB group rats adipose tissue perilipin protein expression as well gene expression were markedly higher than C group (P<0.05),but significantly lower than OR group(P<0.01).The OB group PPAR-γgene expression was markedly higher than C group(P<0.01).2.Compared with the non-exercise control group(EC group),after a single bout of exercise,the serum FFA and serum glycerol concentrations, adipose tissue HSL activity,HSL protein and gene expression were markedly elevated at different times.However,the adipose tissue perilipin expression was only higher immediately after a single bout of exercise,and markedly lower at other times.The perilipin and PPAR-γgene expression declined significantly at different times,too.3.Compared with the long-term control group(LC group),the weight,serum FFA and serum glycerol concentrations of long-term exercise group(LE group) reduced markedly.The LE group adipose tissue HSL activity markedly rose(P<0.01);however,the LE group adipose tissue HSL protein expression tended to decline,and the adipose tissue perilipin protein expression(P<0.01),HSL(P<0.05),perilipin(P<0.05)and PPAR-γgene expressions(P<0.05) reduced markedly.Conclusions:1.10-week high fat diet result in rats to become obese and dyslipidemia, this may be related to the adipose tissue PPAR-γgene expression elevation to upregulate perilipin gene expression and perilipin protein expression,since perilipin may induce fat storage and obesity;on the other hand,the downregulation of rats adipose tissue HSL activity,HSL protein expression as well as HSL gene expression were also related to the bad results.2.A single bout of exercise may result in the increment of obese rats lipolysis during and after exercise.And the mechanisms of lipolysis rising may be involved in the upregulation of its adipose tissue HSL activity,HSL protein expression,HSL gene expression as well as the downregulation of PPAR-γgene expression. 3.Long-term exercise may reduce the rats weight,and these decline could have relation with the HSL activity increment as well as perilipin protein level and gene level decline,and had relation with the downregulation of the perilipin regulatory factor PPAR-γgene expression,too.
