This dissertation devotes the studies mainly to the field of analytical chemistry and analytical instrumentation, and the key point of the dissertation is the technology development in mass spectrometry and instrumentation development of mass spectrometer for the hot research such as drug and protein biology. Mass spectrometry instrument technology is one of glowing spots about advance science and technology, which is believed to be one of the hardest technology to know well for its interdisciplinary fields involving chemistry, biology, physics, microelectronics, computer science and engineering etc. The innovation of this dissertation are listed as followed:(1) developed modules for tandem mass spectrometry based on the TOF MS technology includes vacuum module, ion transportation module, the electronic board card and software based on labview; designed a new interface for ESIT, made the important section and experiment instrument finally got the patents for this new technology;(2) developed a model for scanning frequency of quadrupoles, got the result of experiment and patents for this method; got many patents for ion source interface and small volume mass spectrometry.The main contribution of this dissertation are listed as followed:(1) the general module of mass spectrometry are the key point for the industry of mass spectrometry, it had important significance to decrease the cost and increase the instruments’ use wide, gave important basis for our country’s own industry of mass spectrometry;(2) the high resolution ESIT interface is the key technology for the mass spectrometry interface, it can increase the effiency of ion transportation, solve the problem of obitrap analyzer, it’s believed to be key point technology of super resolution mass spectrometry in the next10years;(3) the scanning frequency of quadrupoles are the new method had important meaning, it can enlarge the quadrupole’s use wide and provided guiding meanings for the improvement of quadrupoles used in the analysis for proteomics.This doctoral dissertation consists of five parts and the contents are summarized as follows:In the first chapter, it mainly introduces the instrument of mass spectrometry, especially about the TOF-MS instrument’s principles and developments. As the development of science and technology, the required for the mass spectrometry which is used for biology is higher than ever, I combined our research basis and gave the working highlights and research direction of this dissertation. Mass spectrometry analysis was ionized the samples which are introduced to the ion source and guided the ions to the mass spectrometry analyzer, the ions separated according to their different m/z, and gave the spectrometry at last. The ion source and analyzer are different for the different mass spectrometry, MALDI and ESI are the main ion source for the biology samples like genomics and proteomics, the TOF or TOF related analyzer has the important status in the development of mass spectrometry, so it’s important to research the TOF-MS mass spectrometry and related technology.The research works describes in the second chapters centers on our own developed ESI-TOF MS instrument. LC-MS are the basis technology for the bio-mass spectrometry analysis, the interface technology is the key point of LC-MS, we choose the ESI to be the ion source of our own instrument and design this ESI-Oa-TOF-MS. We used three different quadrupoles in this instrument and positive/negative pulse mode for the ion introduce. The reflector had second net, the ions detection technology and the MCP position also contained high innovation. The TDC system used for signal detection was simple for the user. The resolution of this instrument is about10000-15000after debugging. In the second part we also introduced the work we already done about the1.2meter TOF-MS and the0.6meter TOF-MS instrument, generalized the experience and shortage also gave the main effort in future.For the works mentioned in the third chapter, the table-type ESI-Oa-TOF-MS instrument for the industry used is well tried and established. The Time of Flight analyzer was designed with the homogeneous field which is international band technology, also used three different quadrupoles, decreased spatial scattering and energy scattering of the ion beam effectively, the reflector and ion detection method are the same with the former instrument. The character of this instrument was well coupled with the LC made in china and it’s very cheap fit for our country’s market.In the fourth chapter, the development of modules for some kinds of mass spectrometry is mentioned. The universal modules for mass spectrometry has important significance in the development of mass spectrometry instrument, it can simplify the step of designing and decrease the cost for development and manufacture. The universal modules we designed consist of ion transport system, electronic system, the key section of mass spectrometry like quadrupoles, TOF analyzer, the system for test and the electronic board,the software which is based on labview and CVI also the vacuum module. All the modules have Chinese intellectual property rights, it brought the basis for the industry development of mass spectrometry in china.In the last chapter, developments on other type mass spectrometry are summarized. The main technology concentrated on the quadrupoles analyzer, ion trap and ESIT analyzer. The principles of every kinds of analyzer was well analyzed, the process technology and metrology system also mentioned in this part, the important technology was well explained, the quadrupoles mass spectrometry which used for industry was also mentioned, these work provided high referential value for the development of mass spectrometry in china.
