Objective to investigate cytokine expression in the intestinal mucosa from patients with inflammatory bowel disease by antibody chips, elucidate the role of the screened proteins in pathogenesis of inflammatory bowel disease,and validate the reliability of the screened proteins by antibody chips using Western blot.Methods to collect the fresh mucosa of9patients with ulcerative colitis,9patients with Crohn’s Disease and9healthy individuals, extract total protein of each group, and compare different cytokine expression of3groups by Raybiotech human L-series cytokine antibody chips. To validate the greatly differential expressed proteins by Western blot.Results comparing with the healthy controls, the levels of263cytokines changed significantly in UC, and the levels of414cytokines changed significantly in CD. They were GDNF family receptors a,Chemokines, Matrix Metalloproteinases and its inhibitors, Fibroblast growth factor, Leukemia inhibitory factor, Interleukin, Transforming growth factor and Smad proteins, Calcium-binding protein S100A12,Leptin,Pentraxin3, Adhesion molecules, Vascular endothelial growth factor and its receptors, Tumor necrosis factor superfamily and their receptors. Herpesvirus entry mediator, Leukemia inhibitory factor and platelet factor4were validated by Western blotConclusion the differential cytokine expression in IBD can help to discover the pathogenesis of IBD and search some new biomarkers.
