BackgroundCoronary artery ectasia (CAE) is defined as localized or diffuse dilatation of coronary artery lumen exceeding the largest diameter of an adjacent normal vessel more than1.5fold.The gold standard for diagnosis CAE is coronary Angiography,and the Corrected Thrombolysis in myocardial infarction frame count(CTFC) is a simple clinical tool for assessing quantitative indexes of coronary blood flow.GDF-15is a distant member of the TGF-β cytokine superfamily. High level of GDF-15is induced by various forms of stress, and it plays an protective role in the pathological process of cardiovascular diseases.. Single Nucleotide Polymorphisms (SNP) are found to be associated with the bioactivity of GDF-15.To our knowledge it is unknown the relationship between the plasma level and genotypes of GDF-15and CAE, this research aims to investigate if GDF-15and its genotype posses some diagnostic and prognostic values in patients with CAE.MethodsBlood samples were obtained before the coronary angiography (CAG) from47patients with CAE and30patients with normal coronary artery. GDF-15genotypes were determined by direct sequencing method. Review the CAG results to determine the CTFC. Clinical and biochemical information was recorded. Sequence GDF-15genotype in three SNP loci rs1059519, rs1059369and rs1058587. Statistical analysis was performed by SPSS16.0. ResultIn patients with CAE, the maximum CTFC and GDF-15level are positively correlated, the regression equation:Y=22.94X1+98.81. Regression correlation coefficient R2=0.561. The genotypes of loci rsl059519are associated with GDF-15plasma level, CC, CG, GG patients with type GDF-15plasma levels were elevated (812.5±383.1,1191.5±449.9,1390.8±597.4,P=0.026). Divide the CAE patients in to two subgroups by the maximum CTFC above30and below. The frequencies of allele C or G of loci rs1059519are different between the two subgroups. The frequencies of allele G in the blood flow restricted group is higher, OR=4.490,(95%CI,1.113-15.60), P=0.034.Multiple regression analysis shows that CTFC and GDF-15loci rs1059519genotypes were associated with the expression of GDF-15, Y=-153.6+285.9X1+19.61X2. Regression correlation coefficient R2=0.518.The partial correlation analysis showed that the introduction of GDF-15genotype does not affect the relationship of GDF-15levels and coronary dilation, standards relevant coefficient (0.675vs0.647).ConclusionsIn patients with CAE, GDF-15is related to coronary artery perfusion. GDF-15genotypes are associated with GDF-15plasma level and severity of coronary artery ectasia significantly.
