Objective:Chronic fatigue syndrome (CFS) is a systemic syndrome characterized by sustained fatigue and activity reduction whose duration is more than or equal to six months, accompanied by sore throat, headache, muscle and joint pain, lymph node enlargement and tenderness, fever (or consciously fever), sleep disorders, poor memory, depression and a variety of neuropsychiatric symptoms, could not be released by bed rest, yet any physical examination and laboratory examination show the obviously abnormality. But at present, the etiology and pathogenesis of the disease have not been studied thoroughly at home and abroad, which may be associated with viral infections, neuroendocrine disorders, abnormal autoimmune function, genetic and other factors. The study uses animal experiment to discuss the effect of comprehensive syndrome rats back-shu acupoint catgut embedding therapy on patients with chronic fatigue, for finding the mechanism of its effect, and to provide the theoretical basis for clinical application of this therapy.Methods:This research uses 40 SPF grade SD rats which was 3 month’s age,160-180g body weight, each half of the sex. Those were divided into blank group, model group, catgut embedding group and drug group by random digit table method randomly. There were 5 male and female rats in each group, and they were adapted to 2 days after modeling while the sum was 10.The CFS rat model was prepared by chronic restraint and forced cold water swimming method, except the blank control group, bounded at singular date, while swimming in cold water at double date, lasting 14 days. The rats in the blank group were reared in the same environment, and were suspended for 1 times daily, and the blank group was not fed and fed during the rest of the model. Chronic restraint:the three groups of rats in the model were bound to be bound for 1 time each day, and the time of each bound was different, randomly determined. When they were bounded, the rats were drilled into the restraint cylinder (and was adjusted to the tension level that does not produce a strong resistance, and was flat on the board. After 6 minutes it was removed. Forced swimming in the cold water method:prepare two swimming pools and specifications for 185cm*58cm*73cm, swimming depth of 50cm, the water temperature for 14+L C, with groups of male and female rats also were placed in two swimming pools,8 minutes later removed. Prevent the rat group to save themselves or due to the exhaustion of the water during the swimming.1 hour within the daily model treatment after treatment,1 times a day, for 14 days. The blank group and the model group were treated with no treatment.The prescription selected in the embedding group was as follow:first group: Feishu combine Xinshu; the second groups:Ganshu, Pishu and Shenshu. Catgut embedding therapy was set for next day, two groups by alternating points. The drug group was treated with 7mg/100g (Ginsenoside 70mg/kg/d), and the standard of daily administration of ginsenoside aqueous solution was administered to the stomach for the treatment of each rat. The two groups were treated for 14 days, and each treatment was performed at 1 hour after the treatment.Including the amount of drinking water, food intake, body weight and rat feces, the hair color and squinting, ear color, tail thickness and color, when the rest of the body posture, every day of the general condition of the rats were observed. At the end of the experiment, detected the behavior science (open field test and suspension experiment), spleen index, serum IL-1, IL-6, cerebral cortex BDNF and NGF expression levels.Results:1. Before set up the model, there were no significant differences between the rats. With the time of modeling, the rats in the blank group were quiet or active, smooth fur, eyes fresh; Rats in the model group were a slight decrease in eating and drinking, burnout, fur lose its natural luster, eyes dim, thin, reduced movement, squint, stool thinning, love together, the reaction force decreased obviously. Rats in the treatment group mental health is fine, eating and drinking more than the model group, fur、activity and faeces were no significant differences of blank group.2. After the building the model and treatment, the model compared to the blank shows, The number of horizontal movement and vertical movement of rats decreased significantly(P<0.01); the catgut implantation at acupoint and drug compared to model shows, The number of horizontal movement and vertical movement of rats increased substantially, The differences were statistically significant(P<0.05).3. Rats in the model group tail suspension test were struggling times becomes diminished, still time more than the other group, it compared to blank group, the differences were statistically significant(P<0.01);the catgut implantation at acupoint and drug compared to model group the still time becomes diminished, the differences were statistically significant(P<0.05).4. The spleen coefficient of rats in model group were significant lower than blank group, the differences were statistically significant(P<0.01); After the catgut implantation at acupoint and panaxoside, the spleen coefficient of rats were rising, the differences were statistically significant with model group(P<0.05), among the catgut implantation at acupoint spleen coefficient of rats were significant rising(P<0.01).5. The IL-1β、IL-6 in model group were significant riser than blank group, the differences were statistically significant(P<0.01); The IL-1β、IL-6 in the catgut implantation at acupoint and drug group were significant lower than model group, the differences were statistically significant(P<0.05), among IL-1β、IL-6 in the catgut implantation at acupoint group decreasing amplitude maximum (P<0.01).6. The optical density and positive area in model group were significant riser than blank group, the differences were statistically significant, it was showed that the cerebral cortex’s BDNF、NGF in the model group were significant better than blank group. the cerebral cortex’s BDNF、NGF in the catgut implantation at acupoint and drug group were significant lower than model group, the differences were statistically significant.Conclusions:Selected Five Back-shu Points catgut implantation at acupoint to Chronic fatigue syndrome rats intervention can slow down the model rats of various symptoms of fatigue and weight increased slowly, and improve its behavior, increasing physical fitness and correct the depression negative emotions. Its underlying mechanics may Back-shu Points catgut implantation at acupoint could improve the model rat spleen coefficient, to improve their own immune system, improve the content of neurotrophic factor in brain tissue, to improve the psychological-nerve -immune system, thus the treatment of chronic fatigue syndrome. Catgut implantation at acupoint were similar results have been found with panaxoside.
