| Object:To explore the mechanism of Compound Phellodendron Liquid in treatinginfectious wound,Through observing the curative effect of patients with postoperative analfistula and the expression of epidermal stem cells (ESCs), TNF–α,IL-6, β-catenin andSDF-1α regulation on the rat model of infectious wound.Method:60patients with postoperative anal fistula were Randomly divided into twogroups,30cases treated by Compound Phellodendron Liquid,while30cases treated byrutine methods,.Observing the woud healing. Two full thickness dermal excision woundmodels were established on the back of the Wistar rats,some wound were injectingstaphylococcus aureus and escherichia coli to make infectious wound animalmodel,divided into:normal group, model group,Compound Phellodendron Liquidgroup,and Gaifu group(western medicine control group),The medicine were attach to theskin once a day,to calculate the area of wound surface and healing rate,and after settingmodels for3,7and15days,the rats were killed to draw the wound surface,HE stained toobserve the skin regrowth state, The expression of ESCs surface marker of β1-integrinand k19were compared by IHC, the level of TNF-α(mRNA),IL-6(mRNA), β-catenin(mRNA) and SDF-1α(mRNA)were detect by ELISA and RT-PCR method.Results: Clinical observation showed the case treated by Compound PhellodendronLiquid was much effective at healing rate, healing time and relieving pain compare withcontrol group group. Animal experiment showed that the healing rate of normal group washigher than the model group,the expression of ESCs surface marker of β1-integrin andk19and at the model group was lower than the normal group, while the TNF-α(TNF-α mRNA) and IL-6(mRNA) was more higher,treated by Compound Phellodendron Liquid,the expression of TNF-α(mRNA) and IL-6(mRNA) was lowered,and the level of β-catenin(mRNA) and SDF-1α(mRNA)were higher,and enhance the expression of ESCssurface marker of β1-integrin and k19.Conclutions: High levels of TNF-αand IL-6can delayed the wound healing time andpossibly relate with inhibiting epidermal stem cell proliferation, Compound PhellodendronFluid can promoting the infectious wound healing, one of its mechanisms may by loweringthe level of TNF-αand IL-6, improving wound inflammation pathological state, and thenactivate β-catenin which was the key factor relate with proliferation and differentiation ofepidermal stem cell, enhance the expression of chemotactic factor SDF-1α, soproliferation and migration of epidermal stem cell in edge of skin wound surface waspromoted. |
