| Object To compare the bacteriostatic effect of different extraction schemes of compound hibiscus mosaic in vitro,and to provide a basis for further study of compound hibiscus mosaic.Next to explore the mechanism of compound hibiscus mosaic extract on wound healing in rats.Method First,ethanol,ethyl acetate and water solvents were used to extract the compound hibiscus flower leaves,and then the extracts were diluted twice to inhibit Escherichia coli and Staphylococcus epidermidis in vitro.Then 60 rats were selected and divided into blank group(n=15),model group(n=15),gentamicin group(n=15),compound hibiscus flower leaf group(n=15)according to the random number table method.After modeling the experimental rats,observe the wounds of the rats in each group at 3 days,7 days,and 15 days after the intervention,and measure the size of the wound diameters of the rats in each group at 3 days,7 days,and15 days after the intervention.And use HE staining on the wound tissue to observe the pathological changes of the wound tissue.The wound tissue was collected and used ELISA to detect the levels of interleukin-1 and acidic fibroblast growth factor.Result 1.The extraction rate of ethanol extraction was 26.60 %,the minimum inhibitory concentration of E.coli was 68.18 g/L,and the minimum inhibitory concentration of Staphylococcus epidermis was 34.09g/L.The extraction rate of ethyl acetate extraction was 1.38 %,the minimum inhibitory concentration for E.coli was 4.26 g/L,and the minimum inhibitory concentration for Staphylococcus epidermis was 1.07g/L.The extraction rate of water extraction program was 30.40 %,the minimum inhibitory concentration of E.coli was 68.18 g/L,and the minimum inhibitory concentration of Staphylococcus epidermis was 4.26g/L;2.In comparison of the diameter of the healing wound,the diameter of the wound in the compound hibiscus mosaic group was the smallest(P<0.05)and healed best at 7 and 15 days after the intervention(P<0.05);compared with the pathological section of the wound,the compound hibiscus mosaic group Compared with the other groups,the pathological sections of the group had fewer inflammatory cells,hyperplasia of granulation tissue,and increased myoblasts.In the ELISA test results,the level of interleukin-1 in the compound hibiscus mosaic group was significantly lower than that in the model group and gentamicin group(P<0.05)after the intervention.The level of interleukin-1 in the gentamicin group was always higher than that in the other groups.Seven days after the intervention,the content of fibroblast growth factor in the compound hibiscus flower and leaf group was higher than that of the other three groups.Conclusion 1.Regardless of the extraction method of compound hibiscus flower leaves,it can produce obvious antibacterial effects on E.coli and Staphylococcus epidermis.The optimal extraction method for obtaining the lowest antibacterial concentration is ethyl acetate extraction.2.The compound hibiscus mosaic controls the level of interleukin-1and up-regulates the expression of acidic fibroblast growth factor in the wound tissue,so as to control the inflammation of the wound,reduce inflammatory cells and edema,promote the proliferation of granulation tissue,and accelerate the wound healing time. |
