Bidouyan-oral Liquid For Acute Nasal Sinusitis Model The Effect Of EGF And EGFR Expression In The Rat Nasal Mucosa

Objective:The research We observe the effect of Bidouyan-oral solution on beta-defensin in nasal mucosa of ARS rats modal, meanwhile, studythe mechanism of action about it to Bi Yuan by animal experiment, then to guide the clinical medication more accurately. Methods:Group the healthy SD rats into the following six parts randomly: blankgroup, model group, Ji Lei Kai Qiao Tang low-dose group, Ji Lei Kai Qiao Tang middle-dose group, Ji Lei Kai Qiao Tang high-dose group and Amoxicillin and Clavulanate Potassium group. According to the method reported by YE Jing, we madethe Acute Rhinosinusitis model.In the following seven days we drenched the blankgroup and the model group 5ml·kg-1 d-1 with the sterile water (SW) for 7 days,meanwhile the other four groups with corresponding medicine. All these days weobserved the changes of the rats about the pH value, body temperature, and bodyweight, especially the nose symptom and secretory volume. After seven days,6 ratsrandomly choosed in each group to exam ph of nasal secretions, leucocytecount(WBC) and the percent of neutrophils (N%)of peripheral blood cells,then wekilled the rats to get nasal mucosa to observe the morphologic changes of it underlight microscope. AT the time,SP immunohistochemistry method was used to testEGF.EGFR in nasal mucosa, and RT-PCR(real time polymerase chainreaction) methods was used to test EGF mRNA.Results:1.Comparing with the blank group, the PH value of the model group’s nasal cavity secretion substance has improved significantly (P<0.01). The nasal cavity secretion substance’s pH values of all the therapy groups have declined, especially the B idouyan-oral liquid high-dose group and Amoxicillin and Clavulanate Potassium group.2.2.Comparing with the blank group, WBC and N% of the model group has improved significantly (P<0.01). WBC of all the therapy groups have declined significantly (P< 0.01), especially the B idouyan-oral liquid high-dose group and Amoxicillin and Clavulanate Potassium group.3.Compared with the blank group, model group in the rat nasal mucosa tissue EGF immunohistochemical detection integral optical density (MD) decreased significantly (P< 0.05) Compared with model group, Western medicine group, high dose group and the dose in the way of rat nasal mucosa tissue EGF im munohistochemical detection integral optical density (MD) were significantly hi gher (P< 0.05), low dose group has increased.4.Compared with the blank group, model group in the rat nasal mucosa tissue EGFR immunohistochemical detection integral optical density (MD) significan tly lower (P< 0.01), suggesting building success. Compared with model group, Western medicine group, high dose group and middle dose group of rat nasal mucosa tissue EGFR immunohistochemical detection integral optical density (MD) were significantly increased (P< 0.01), low dose group has increased.5.Compared with the blank group, model group in the rat nasal mucosa tissue EGF gene expression is significantly lower (P< 0.01). Compared with model group, Western medicine group, high dose group of rat nasal mucosa tissue E GF gene expression is significantly higher (P< 0.01); Dose groups in rat nasal mucosa tissue EGF gene expression is significantly increased (P< 0.05).6.Using the light microscope,Significant pathological changes of acute inflammation were observed in nasal membrane of the model group.After seven days medication,except the Ji Lei Kai Qiao Tang low-dose group.the nasal membrane of all the therapy groups have returned to normal gradually,especially the B idouyan-oral liquid high-dose group Amoxicillin and Clavulanate Potassium group.Conclusions:1.By falling the Nasal discharge pH value to normal, Bidouyan-oral liquid can make the internal environment concordance of the nasal cavity.2.Groups of nasal mucosa of rats between EGF MOD values have significant difference (P< 0.05), EGF is inherent expression in rat nasal mucosa. Wester n medicine group, the Bidouyan-oral liquid high dose group, middle dose grou p can enhance the ARS rat nasal mucosa EGF gene expression, give play to t he role of repair, effectively promote the sinus mucosa wound healing.3.Each rat nasal mucosa between EGFR MOD values have extremely signific ant difference (P< 0.01), EGFR is inherent in the nasal mucosa of rats. EGFR can be induced under inflammatory conditions. Bidouyan-oral liquid high dos e group, medium dose group, amoxicillin/Clavulanate Potassium can effectivel y control the local inflammation, strengthen the nasal mucosa tissue repair an d functional recovery.4. B idouyan-oral liquid can control the general and local inflammatory reaction in ARS rats, especially the high dose.
