Eighteen antagonisms is obtained from physicians clinical practice experience over millennia as the incompatibility in TCM. Although it has been used in TCM, its scientific connotation is still not clearly defined. The incompatibility mechanism is generally recognized as a serious side effect or toxic reaction. But there is also a kind of understanding that incompatibility can interfere, change and reduce or eliminate the efficacy, but the universality and science of obstruction treatment phenomenon remains to be further scientific validation.In this paper, we mainly studied on the mechanism of Panax ginseng incompatible with Veratrum nigrum in pathological statethe as the representative of eighteen antagonisms. First of all, the untargeted metabonomics method based on UPLC/Q-TOF-MS was established. The biological mechanism of Qi deficiency syndrome and Estrogen deline, the mechanism of action with Panax ginseng, the mechanism incompatibility of Panax ginseng with Veratrum nigrumin were investigated basing on the method before. The main research content as follow:1. The study on screening of different proportion of Panax ginseng and Veratrum nigrumThe effective dose of ginseng had beed pre-screened and fixed. The results of the DPPH radical scavenging,-OH radicals scavenging in vitro and normothermic mice swimming, often resistant to hypoxia in vivo were used to screening the proper proportion of Panax ginseng and Veratrum nigrum. And four proportion was ultimately determined to study the mechanism of Panax ginseng incompatible with Veratrum nigrum. It’s 1:0.1, 1:0.05, 1:0.02 and 1:0.01.2. Untargeted metabonomics study of Qi deficiency syndromeThe biological metabolism caused by Qi deficiency syndrome and the mechanism was detected with the methods of metabonomics based on UPLC-Q-TOF-MS. The 22 endogenous biological markers of Qi deficiency syndrome was identified. And these markers in Tryptophan Metabolism, Citric Acid Cycle, Taurine and Hypotaurine Metabolism, Pantothenate and Co A biosynthesis etc.. By comparing the literatures and biological databases, the changes of the markers are mainly influenced by the Qi deficiency which mainly affect the regulation of immune function, energy metabolism and redox reaction. The results of this study provide clearly biological mechanism of Qi deficiency syndrome. It also indicates that metabonomics is a very good application in the study on the mechanism of TCM syndromes.3. The effect of Panax ginseng incompatible with Veratrum nigrum on Qi deficiency syndromeThe 15 biomarkers for ginseng on treatment of Qi deficiency syndrome were identified. These markers showed that ginseng adjusted Tryptophan Metabolism,Beta-Alanine Metabolism, Citric Acid Cycle, Taurine and Hypotaurine Metabolism etc.. caused by Qi deficiency syndrom.The different proportions of Panax ginseng incompatible with Veratrum nigrum were studied. The results of weight, exhaustive swimming time, blood index and serum biochemical efficacy showed that ginseng had a better therapeutic effect on Qi deficiency syndrome by enhancing energy metabolism, regulating immune function and antioxidant. Compatibility with Veratrum nigrum in different proportions were lower than that of the single ginseng, which was minimal impact in group of 1:0.01 and greatest impact in group of 1:0.1. PCA analysis and the relative content of different biomarkers revealed that the regulation ability of incompatibility on biomarkers is much lower than that of ginseng. The lower proportion of Veratrum nigrum, the smaller effect on treatment of ginseng.4. Untargeted metabonomic study of Estrogen delineThe biological metabolism caused by Estrogen deline and the mechanism was detected with the methods of metabonomics based on UPLC-Q-TOF-MS. The 23 endogenous biological markers of Estrogen deline was identified. And these markers in Pantothenate and Co A biosynthesis, Citrate cycle, Steroid hormone biosynthesis,Valine, leucine and isoleucine biosynthesis etc.. By comparing the literatures and biological databases, the changes of the markers are mainly influenced by the Estrogen deline which mainly affect the regulation of immune function regulation,energy metabolism, fatty acid metabolism, amino acid metabolism and steroidogenesis. The results of this study provide clearly biological mechanism of Estrogen deline and reference for other related research.5. The effect of Panax ginseng incompatible with Veratrum nigrum on Estrogen delineThe 15 biomarkers for ginseng on treatment of Estrogen deline were identified. These markers showed that ginseng adjusted Tryptophan Metabolism, Fatty Acid Biosynthesis, Citric Acid Cycle, Steroid hormone metabolism etc.. caused by Estrogen deline.The different proportions of Panax ginseng incompatible with Veratrum nigrum were studied. The results of index weight, gonad index, rat estrous cycle changes and serum estrogen level showed that ginseng had a better therapeutic effect on Estrogen deline by enhancing energy metabolism, fatty acid metabolism, amino acid metabolism,regulate immune function and anti oxidative. Ginseng had estrogenic effects, but not as the supplements of estroge. Compatibility with Veratrum nigrum in different proportions were lower than that of that ginseng, which was minimal impact in group of 1:0.01 and greatest impact in group of 1:0.01. The lower proportion of Veratrum nigrum, the smaller effect on treatment of ginseng.
