Objective:The purpose of this paper is to explore the expression and significance of BMP-4,5,6,7 in rats with traumatic brain injury combined fracture, to investigate the mechanisms of traumatic brain injury promoting fracture healing, and provide the theoretical basis for treatment of fracture, delayed union and ununion.Method:110 healthy male Standard rats were randomly divided into three groups:Group A (Blank control group), Group B (Fracture group)、Group C (Fracture combined traumatic brain injury group).group A was 10 rats and B, C groups each 50. Group B established Tibia fracture model and Group C established Moderate traumatic brain injury model. Observed the general state of the two groups of rats, including animal body weight, mental condition, activity, food intake, water quantity, etc. Observed the changes of damaged brain tissue under optical microscope at the time of 1,2,3,5,7 weeks after the experiment in group C. Observed fracture healing through the X-ray at the time of 1,2,3,5,7 weeks after operation in group B and C. Measured the Callus diameter at the time of 3,5,7 weeks after operation in group B and C. Observed the changes of hematoma and callus tissue under optical microscope at the time of 1,2,3,5,7 weeks after the experiment in group B and C. measured the mRNA and protein of BMP-4、5、6、7 in serum and callus at the time of 1,2,3, 5,7 weeks after the experiment in group A, B and C. Analysis data of the experiment at last.Results:It was found that activities of the rat in group C increase gradually since 2 weeks after operation, and resume normal activities at the time of 4 weeks or so after operation, earlier than the group B; Through optical microscope It was found that Brain tissue is injury and bleed in the early, then appear empty, necrosis. Through X-ray It was found that some callus appear around the fracture of rat at the time of 2 weeks after operation in group C, a lot of callus can be seen and the fracture line is indistinct at the time of 3 weeks after operation, The fracture line disappear almost at the time of 5,7 weeks after operation. It was found the X-ray density of callus in group C is higher than that in group B(P<0.05).The callus diameter of rats in group C was greater than that in group B(P<0.05).In the early a lot of new blood vessels and bone trabecula can be seen in hematoma and callus around the fracture in group C. The time of Callus formation and remodeling in group C was earlier than that in group B(P <0.05).The serum BMP-4,6,7 gene mRNA of the rats in two groups showed a high level of expression and changed with time (P< 0.05).The serum BMP-5 gene mRNA of the rats in two groups showed no abnormal expression. The BMP-4,6,7 gene mRNA in serum and callus of the rats in group C were higher than that in group B(P<0.05). The serum levels of BMP-4,6 and 7 protein were high in the two groups of rats and changed with time (P<0.05).The serum level of BMP-5 protein was not abnormal. The levels of BMP-4,6,7 in serum and callus of the rats in group C were higher than that in group B(P<0.05).Conclusion1. Moderate traumatic brain injury can promote fracture healing.2. BMP-4,6,7 may be one of the factors that promote the healing of the fracture. BMP-5 does not participate in the regulation of traumatic brain injury in promoting fracture healing.3.BMP regulatory mechanism in the process of gene transcription and translation may be similar...
