Objective: Through the experiment research to Expression of Different time points, In the serum and partial callus, Bone morphogenetic protein-7(BMP-7) 、 Transforming growth factor-β1(TFF-β1) during Tibial fracture healing combined traumatic brain injuries in Standard rat.TO Preliminary discussion on its accelerate fracture healing effect.And provide new theoretical basis for clinical accelerate fracture healing.Methods : 96 healthy male Standard rats were randomly divided into four groups: Group A(Fracture group)、Group B(Fracture combined traumatic brain injuries group、Group C(Brain trauma group)、Group D(Blank control group). A, B, C groups each 30, D group only 6. References to establish Tibia fracture model and Moderate brain injury model. Extraction of abdominal venous blood before operation(0 d),postoperative 3 d, 1 w, 2 w, 3 w, 4 w. Determination of BMP- 7 and TFF-β1content in serum use ELISA Detection method. Group A and group B respectively after the success of the mould 3 d, 1 w, 2 w, 3 w, 4 w executed respectively 5 only for X-ray radiography, observation of fracture healing. And at the tibial fracture broken end take the callus specimen. For HE dyed observe the fracture healing process. Expression of BMP-7 and TFF-β1 in Partial callus use Immunohistochemical detection method on Different time points. And will appeal against the results of statistical analysis.Results: 1、Macroscopic observation and X-ray radiography showed:Fracture combined traumatic brain injuries group comeblined with Fracture group,Obviously the fracture healing process accelerated,have more callus In fracture, Even the phenomenon of heterotopic ossification.2 、 Expression of BMP-7 and TFF-β1 In the serum:Four groups of BMP- 7 and TFF-β1 the Content of each other of 0 d had no statistical significance(p > o. 05)。Compared with the blank group After building 3d、1w、2w、3w、4w, The rest of the other three groups the Content expression of BMP- 7 and TFF-β1were improved obviously.For BMP – 7:Compared with B and Cgroup The 3 d content is slightly higher, Peak in advance. Group of differences between the same time piont have statistically significant(P < 0.05). Over time Group B Conpared with group A have more contend increased obviously. The same point in time the difference between the two groups content have statistical significance.For TGF-β1: Group B compared with group A, 3 d, 1 w, 2 w expression quantity increased significantly. In advance, The peak(P < 0.05) Between A, B, C three group at the same time points(3 d- 4 w) content differences were statistically significant(p < 0.05).3、HE dyed show:Compared with group A,Group B have more number of fibroblasts, much new blood vessels, distribution, active ossification faster and significantly accelerate the fracture healing process. 4、Immunohistochemical study showed:In the whole process of bone healing, BMP- 7 and TGF-β1 In inflammatory cells, fibroblasts, cartilage cells and expression in osteoblast, with cytoplasmic expression is given priority to, part is also expressed in the nucleus, Group B express more obvious. The expression of BMP 7:After the group B in addition to building 3 d content is lower than that of group A. Were higher than in group A,(1w-4w).3 w and 4 w group contrast between the most obvious, 3 w to 4.22 times in group A, 4 w for group A 5.67 times. Group A and group B in 3 d after injury, 1 w, 2 w, 3 w, 4 w fracture site BMP-7 positive cell percentage of content between the difference had statistical significance(p < 0.05). The expression of TGF-β1: Group B compared with group A, 3 d, 1 w, 2 w content significantly increased and the peak in advance, which is most obvious when 1 w group A content of 2.56 times, 3 w, 4 w is lower than group B, group A and group B in 3 d after injury, 1 w, 2 w, 3 w, 4 w fracture site TGF- beta 1 positive cell percentage of content between the difference had statistical significance(p < 0.05).Conclusion:1、After Tibial fracture merge moderate brain injury in rats compared with the simple tibial fractures fracture healing of rats significantly faster, show that Head trauma on fracture healing.2、Experiments found that fracture combined traumatic brain injuries from fracture group in serum and partial callus significantly higher, the expression of BMP- 7 and TGF-β1. Confirmed the BMP- 7 and TGF- beta 1 is to accelerate fracture healing after fracture combined traumatic brain injuries of impact factor.3、Brain injury when fracture healing to speed up the process of which require the participation of a variety of cytokines. BMP- 7 and TGF- beta 1 is likely to be one of them play a role of the important factors.
