Chinese herbal prescriptions can enhance the clinical curative effect through the use of multi-composition formulas. The combined groups of active ingredients of traditional Chinese medicine with clearer ingredients and mechanisms of action, more stable and controllable product quality is all enhanced mode of formulae organization, which has become a new mode of developing combined components from effective traditional formulas. To obtain the compatibility proportion of groups of active ingredients, methodology of optimizing the ratio among the groups is the most critical. The purpose of this study is that, studying the best ratio among groups of active ingredients of Salvia miltiorrhiza, Panax notoginseng and Herba Epimedii. In the first place, preparing the active ingredients group using modern separation technology, and then, using mathematical statistics combined with modern methods of experimental design optimized the best ratio and dose among groups. This new mode will promote the academic research and the industry development of traditional Chinese medicine.In this paper, the following contents are included:1. To study the optimal technical condition of extracting the total ketone from Salvia miltiorrhiza, two methods, such as supercritical CO2 and ethanol extract were compared. The results showed that there was no difference between two methods, but the method of ethanol extract was more simple and convenient, and fitted for industry production. After ethanol extract from Salvia miltiorrhiza was purified by sodium carbonate, the product purity of total ketone was up to 50%.2. To explore the optimal separation of the total salvianolic acid from Salvia miltiorrhiza by macroporous adsorption resin. Nine types of macroporous adsorption were evaluated for separating efficiency by measuring the adsorption ratio, eluting ratio of total salvianolic acid. The results showed that D-101 macroporous adsorption resin had the best separating efficiency. After enrichment and purification with it, the product purity of total salvianolic acid was up to 68.2%.3. Separation and purification of total flavones from Epimedium sagittatum by macroporous adsorption resin with the help of ultrasonic extraction was developed. Nine types of macroporous adsorption were evaluated for separating efficiency by measuring the adsorption ratio, eluting ratio of total flavones. The D-101 macroporous adsorption resin had the best separating efficiency. After enrichment and purification with it, the product purity of total flavones was up to 63.8%.4. Exploring the optimal separation and purification processes of total saponins from Panax notoginseng with eight different types of macroporous adsorption resin by measuring the adsorption ratio, eluting ratio and et al.. With the optimized purification process, the content of total saponins was up to 72%.5. The best ratio among group was optimized by the capability of protecting endothelial cells against damages induced by LPC combined with modern methods of experimental design. Finally, the human umbilical vein endothelial cells (HUVECs) were treated with LPC (10μg/ml) for 24 h to establish the model of endothelial cells injury. Secondly, HUVECs were prior exposed to drug for 1 h, and then exposed to LPC in the presence of the drugs for 24 h in all; And also we set the control group; At the end of the experiment, the cultured medium was collected for measuring the concentration of lactate dehydrogenase(LDH), nitric oxide(NO), maleic dialdehyde(MDA) as well as asymmetric dimethylarginine(ADMA). The result showed that the concentration of LDH(P<0.05), MDA(P<0.05), ADMA(P<0.05) in the medium significantly increased after exposure of endothelial cells to LPC (10μg/ml) for 24 h. Drug can significantly inhibited the elevated concentration of LDH(P<0.05), MDA(P<0.05), ADMA(P<0.05). finally, overall desirability of the capability of protecting endothelial cells was used to evaluate the best ratio among group.6. By observing the different doses of the experimental group on the effects of myocardial ischemia on rabbit blood viscosity and platelet aggregation the best dose was determined..
