A Heavy Load Exercise And Acupuncture Intervention Influence On Rat Skeletal Muscle Mitochondrial Distribution And Function

Objective:The skeletal muscle mitochondria distribution and function were observed after a heavy load exercise. The change of the mitochondrial move and dynamics of proteins were analyzed, the role of these proteins to mitochondria distribution was explored, and the effect of acupuncture intervention on mitochondrial distribution and function was investigated.Methods:SD rats were divided into 4 groups:control, acupuncture, heavy load exercise, acupuncture and exercise. Movement way to run downhill, slope-16°, the speed is 16 m/min,90 min time; Acupuncture method of longitudinal from the distal leg triceps stop at leg triceps muscle belly, angle is about 30°, retaining needle 2 minutes. After the termination of experiments, each group was divided into 0h,12h, 24h,48h and 72h group, and the soleus was detected in the corresponding time points. The transmission electron microscope was used to observe mitochondrial distribution in exercise immediately in each group at different time points. Western Blot was used to detect mitochondrial function protein(SDHA, Cyt C), mitochondrial move protein (Miro1, Miro2, KIF5B, VDAC1, Dynlt1) and mitochondrial dynamics protein (Mfn2, Drp1) expression after a heavy load exercise and acupuncture intervention in skeletal muscle.Results:(1) The numbers and sizes of subsarcolemmal mitochondria were increased in group of heavy load exercise, acupuncture and exercise. Acupuncture group SDHA protein expression increased. Exercise group SDHA protein expression decreased except 12 hours, and there was significant difference compared with the quiet group. Acupuncture and exercise group SDHA protein expression increase in 12,48,72 hours. Cyt C protein expression was up-regulated. (2) Acupuncture group Mirol protein expression appeared decline in 6 hours and 48 hours; Miro2,KIF5B, VDAC1 and Dynltl protein expression increased. Mirol, Miro2, VDAC1 and Dynltl protein expression increased in group exercise and acupuncture combined exercise, but KIF5B change is not obvious. (3) Acupuncture group Mfn2 protein expression increased. Drpl protein expression increased in 12 hours. Exercise group Drpl protein expression decreased in 0 hours. Acupuncture and exercise group Mfn2 and Drpl protein expression had significant differences compared with exercise group.Conclusion:(1) The mitochondria appeared redistribution in group of acupuncture, a heavy load exercise, acupuncture and exercise. Mitochondrial function was decreased by a heavy load exercise while acupuncture can improve it. (2) The subsarcolemmal mitochondria accumulated was associated with mitochondrial move proteins expression changed.(3) Mitochondrial fusion was inhibited after a heavy load exercise, and acupuncture could ameliorate dynamics state, and affected the mitochondria distribution and function.
