Objective:according to the changes of microtubules and mitochondria in skeletal muscle affected by the eccentric exercise and acupuncture, we try to analyze the relationship both microtubule associated protein and the mitochondrial membrane protein, explore whether microtubule could affect the morphology and function of mitochondria induced by the eccentric exercise and acupuncture, and enrich the possible mechanisms of exercise-induced skeletal muscle injury and acupuncture repair.Methods:SD rats were divided into 4groups:control, eccentric exercise, acupuncture and sports acupuncture. Western Blot was used to detect both tubulin(including α-tubulin and MAP4) protein expression and mitochondrial function protein(VDAC, Cyt C and COXI) expression affected by 3-week-exercise and a bout of exercise. Co-IP was used to detect the combination of MAP4 and VDAC. The transmission electron microscope was used to observe mitochondrial ultrastructure. In addition, C2C12 cells were treated by the microtubule depolymerization, microtubule polymerization and electrical stimulation Respectively, and detected the relative Test indicators by Western Blot and Co-IP.Results:There was not significant changes in α-tubulin protein expression, and MAP4 protein expression decreased after 3-week eccentric exercise. The protein expression of VDAC and Cyt C increased, and COX I protein expression decreased. Exercise and acupuncture could improve the combination of MAP4 and VDAC. The intervention of acupuncture could weaken the changes of MAP4 and mitochondrial functional protein expressions induced by eccentric exercise. After a bout of eccentric exercise, α-tubulin protein expression decreased, MAP4 expression increased. Mitochondrial distribution and morphology appeared the typical ultrastructure pathology changes. The expressions of VDAC and Cyt C proteins increased, and COXI expression decreased. The combination of MAP4 and VDAC significantly increased after 12h,24h and 48h. The changes of every protein expression were declined after intervention of acupuncture, α-tubulin protein expression of C2C12 cells declined, MAP4 expression increased, and the expressions of VDAC and Cyt C also increased after the treatment of colchicine and electrical stimulation. The expressions of α-tubulin, MAP4 and COXI increased after the treatment of taxol. The changes of α-tubulin, MAP4 and Cyt C expression were weaken after both electrical stimulation and taxol.Conclusion:eccentric exercise could induce depolymerization of microtubules, microtubule could affect mitochondrial morphology and function by the combination of MAP4 and VDAC. Acupuncture intervention could not only ease microtubule depolymerization, but also could can also alleviate the damage of mitochondrial morphology and function by the MAP4-VDAC way.
